補腎填精方對去卵巢骨質疏松模型大鼠膠原代謝的影響

【摘要】:目的通過對比觀察補腎填精方左歸丸和左歸丸去補陽藥對去卵巢骨質疏松模型大鼠膠原代謝的影響,探討左歸丸陽中求陰法防治骨質疏松癥的作用機制。方法取雌性SD大鼠32只,采用雙側卵巢摘除術建立骨質疏松模型,隨機分為假手術組(Sham)、模型對照組(OVX)、左歸丸組(ZGP)、左歸丸去補陽藥組(ZGPQ),每組8只。灌胃給藥12周后收集各組大鼠血清和股骨組織,采用ELISA法檢測血清中PICP和ICTP水平,采用Masson染色法檢測股骨組織膠原纖維形態(tài),采用免疫組織化學染色法檢測股骨組織ColⅠ蛋白表達水平。結果與Sham比較,OVX血清PICP和ICTP水平顯著升高(P0.01)、股骨組織ColⅠ蛋白表達顯著降低;與OVX比較,補腎填精方下調血清PICP和ICTP水平(P0.05,P0.01)、上調股骨組織ColⅠ蛋白表達。結論補腎填精方左歸丸對去卵巢引起的骨質疏松癥具有一定的防治作用,其作用機制可能與其干預膠原代謝有關,全方效果較為明顯。
[Abstract]:Objective to observe the effects of Zuo Gui Pill and Zuo Gui Pill on Collagen Metabolism in Ovariectomized Ovariectomized Rats, and to explore the mechanism of preventing and treating osteoporosis by Zuo Gui Pill. Methods Osteoporosis model was established in 32 female SD rats by bilateral ovariectomy. The model rats were randomly divided into sham-operated group (OVX), model control group (OVX), Zogui pill group (ZGPGX), Zogui pill group (ZGPQN), each group (8 rats). The serum and femur tissues were collected 12 weeks after intragastric administration, the levels of PICP and ICTP in serum were detected by ELISA method, and the morphology of collagen fibers in femur tissue were detected by Masson staining.
