【摘要】：花生四烯酸經(jīng)過細(xì)胞色素P450(cytochrome P450,CYP)表氧化酶途徑生成環(huán)氧二十碳三烯酸(epoxy eicosatrienoic acid,EETs),具有擴(kuò)張血管、降低血壓、抗炎等多種生物學(xué)功能。在哺乳動(dòng)物系統(tǒng)中的可溶性環(huán)氧化物水解酶(soluble epoxide hydrolase,s EH)具有α/β水解酶折疊結(jié)構(gòu),對(duì)環(huán)氧脂肪酸具有高度的選擇性。s EH能夠快速水解EETs,增加患心血管疾病的風(fēng)險(xiǎn)。目前,研究發(fā)現(xiàn)s EH抑制劑具有抑制s EH活性、提高EETs的含量的重要功能。在多種疾病動(dòng)物模型中應(yīng)用s EH抑制劑或s EH基因敲除,證實(shí)s EH在心肌肥厚、糖尿病、高血壓和腎病等疾病中發(fā)揮重要的生理作用。因此,s EH已被作為疾病治療的新靶點(diǎn)而進(jìn)行研究。本文就s EH的分布、作用機(jī)制以及s EH與疾病的關(guān)系等方面進(jìn)行了討論。
[Abstract]:Arachidonic acid (Arachidonic acid) produces epoxy-eicosyltrienoic acid eicosatrienoic (EETsN) via the cytochrome P450(cytochrome P450 CYPase pathway. Arachidonic acid has many biological functions, such as dilating blood vessels, lowering blood pressure, anti-inflammatory and so on. Soluble epoxide hydrolase (EH) in mammalian system has the folding structure of 偽 / 尾 hydrolase, and has a high selectivity for epoxide fatty acids. S EH can hydrolyze EETs rapidly and increase the risk of cardiovascular disease. At present, it has been found that s EH inhibitors have the important function of inhibiting the activity of s EH and increasing the content of EETs. The use of s EH inhibitor or s EH gene knockout in animal models of various diseases has proved that s EH plays an important physiological role in myocardial hypertrophy, diabetes, hypertension and nephropathy. Therefore, EH has been studied as a new target for disease treatment. In this paper, the distribution, mechanism of action of s EH and the relationship between s EH and disease are discussed.
