五羥色胺受體的免疫調(diào)節(jié)功能研究

【摘要】:五羥色胺(5-hydroxytryptamine,5-HT)又稱血清素(Serotonin),是一種單胺類神經(jīng)調(diào)質(zhì)。近年來,研究發(fā)現(xiàn)免疫系統(tǒng)中也存在5-HT受體及其轉(zhuǎn)運(yùn)體,5-HT通過與其受體相互作用參與調(diào)節(jié)免疫系統(tǒng)的活動(dòng)。本課題旨在探討5-HT受體的免疫調(diào)節(jié)功能,研究主要內(nèi)容包括: 1.采用血凝抑制實(shí)驗(yàn)方法,檢測(cè)不同劑量的5-HT1A受體激動(dòng)劑萘哌地爾和5-HT2A受體拮抗劑富馬酸喹硫平對(duì)雛雞接種苗后新城疫HI抗體滴度的影響。實(shí)驗(yàn)結(jié)果顯示:與21日齡對(duì)照組相比,5-HT1A受體激動(dòng)劑中低劑量組(0.42mg/只/天、0.21mg/只/天)抗體效價(jià)低于對(duì)照組,高劑量組(0.84mg/只/天)高于對(duì)照組,而5-HT2A受體拮抗劑各組(1.64mg/只/天、0.82mg/只/天、0.41mg/只/天)均高于對(duì)照組,但結(jié)果均不顯著。 2.采用MTT法,檢測(cè)5-HT1AR激動(dòng)劑鹽酸釘螺環(huán)酮、拮抗劑鹽酸普萘洛爾和5-HT2AR激動(dòng)劑鹽酸DOI、拮抗劑酮色林對(duì)綿羊外周血T淋巴細(xì)胞體外轉(zhuǎn)化增殖的影響。實(shí)驗(yàn)結(jié)果顯示:5-HT1A受體拮抗劑和5-HT2A受體激動(dòng)劑在10μM至100μM濃度范圍內(nèi)劑量依賴性地抑制ConA刺激T淋巴細(xì)胞增殖;并顯著促進(jìn)靜息狀態(tài)淋巴細(xì)胞增殖活性。表明,5-HT1A受體和5-HT2A受體都參與了淋巴細(xì)胞增殖的調(diào)節(jié),兩者作用相反。 3.采用MTT法,檢測(cè)5-HT1A、5-HT2AR反義寡核苷酸對(duì)綿羊外周血T淋巴細(xì)胞體外轉(zhuǎn)化增殖的影響。實(shí)驗(yàn)結(jié)果顯示:5-HT1AR反義寡核苷酸濃度為0.4μM時(shí)可顯著抑制ConA刺激T淋巴細(xì)胞增殖,5-HT2AR反義寡核苷酸濃度為0.4μM時(shí)可顯著促進(jìn)ConA刺激T淋巴細(xì)胞增殖,而0.4μM的5-HT1AR和5-HT2AR正義寡核苷酸均顯著促進(jìn)ConA刺激T淋巴細(xì)胞增殖。 4.采用放射性免疫方法,檢測(cè)5-HT1AR、5-HT2AR激動(dòng)劑和拮抗劑對(duì)ConA刺激T淋巴細(xì)胞上清液IL-2和IL-4濃度的影響。實(shí)驗(yàn)結(jié)果顯示:不同劑量的5-HT1AR、5-HT2AR激動(dòng)劑和拮抗劑可顯著升高或降低淋巴細(xì)胞分泌IL-2、IL-4量,,無劑量依賴效應(yīng)。 實(shí)驗(yàn)研究的主要結(jié)論如下: 1.5-HT受體可能參與調(diào)節(jié)雛雞體液免疫。 2.5-HT1AR促進(jìn)ConA刺激淋巴細(xì)胞增殖,5-HT2AR抑制ConA刺激淋巴細(xì)胞增殖。 3.抑制5-HT1A或激活5-HT2A受體可增強(qiáng)靜息狀態(tài)淋巴細(xì)胞增殖活性。 4.5-HT1AR、5-HT2AR可能參與調(diào)節(jié)淋巴細(xì)胞分泌IL-2和IL-4。
[Abstract]:Serotonin, also called serotonin, is a monoamine neuromodulator.In recent years, it has been found that 5-HT receptor and its transporter 5-HT are involved in the regulation of immune system by interacting with their receptors.The purpose of this study is to investigate the immunomodulatory function of 5-HT receptor. The main contents of this study are as follows:1.The effects of different doses of 5-HT1A receptor agonist naftopidil and 5-HT2A receptor antagonist quintiapine fumarate on HI antibody titers of Newcastle disease (NDV) after inoculation were studied by hemagglutination inhibition assay.The results showed that the titer of antibody in the low dose group (0.42 mg / rat / day) was lower than that in the control group, and the titer of 0.84 mg / day in the high dose group was higher than that in the control group.The results of 5-HT2A receptor antagonist group were higher than that of the control group (1.64mg / rat / day 0.82 mg / rat / day), but the results were not significant.2.The effects of 5-HT1AR agonist, propranolol hydrochloride, 5-HT2AR agonist doi and antagonist ketoserin on the transformation and proliferation of sheep peripheral blood T lymphocytes in vitro were detected by MTT assay.The results showed that the ConA stimulated T lymphocyte proliferation was inhibited in a dose-dependent manner by 1: 5-HT1A receptor antagonist and 5-HT2A receptor agonist in a dose-dependent manner in the range of 10 渭 M to 100 渭 M, and the proliferative activity of resting lymphocytes was significantly enhanced.It was suggested that both 5-HT 1A receptor and 5-HT2A receptor were involved in the regulation of lymphocyte proliferation, and the effect was opposite.3.The effect of antisense oligodeoxynucleotides (antisense oligodeoxynucleotides) of 5-HT _ 1A and 5-HT _ 2AR on the transformation and proliferation of sheep peripheral blood T lymphocytes in vitro was detected by MTTThe results showed that the antisense oligodeoxynucleotides of 5-HT _ 1AR could significantly inhibit the proliferation of T lymphocytes stimulated by ConA when the concentration of antisense oligonucleotides was 0.4 渭 M. 5-HT _ 2AR antisense oligodeoxynucleotides could significantly promote the proliferation of T lymphocytes stimulated by ConA at the concentration of 0.4 渭 M.5-HT1AR and 5-HT2AR sense oligonucleotides at 0.4 渭 M significantly promoted the proliferation of T lymphocytes stimulated by ConA.4.The effects of 5-HT1AR-5-HT2AR agonists and antagonists on the concentration of IL-2 and IL-4 in the supernatant of T lymphocytes stimulated by ConA were detected by radioimmunoassay.The results showed that different doses of 5-HT _ 1ARN 5-HT _ 2AR agonists and antagonists could significantly increase or decrease the amount of IL-4 secreted by lymphocytes in a dose-dependent manner.The main conclusions of the experimental study are as follows:1.5-HT receptor may be involved in regulating the humoral immunity of chicks.2.5-HT1AR stimulates lymphocyte proliferation stimulated by ConA. 5-HT 2 AR inhibits the proliferation of lymphocytes stimulated by ConA.3.Inhibition of 5-HT1A or activation of 5-HT2A receptor increased the proliferative activity of resting lymphocytes.4.5-HT 1 ARN 5 HT 2 AR may be involved in regulating the secretion of IL-2 and IL 4 by lymphocytes.
