【摘要】：目的探討分析抑制性G蛋白α亞單位Gαi2在大鼠腦缺血再灌注損傷中的表達(dá)變化和對(duì)神經(jīng)細(xì)胞內(nèi)鈣離子濃度和凋亡的影響。 方法將90只SD大鼠隨機(jī)分為假手術(shù)組(SS組，n=30)、腦缺血再灌注組(IR組，n=30)、腦缺血再灌注且腦室內(nèi)百日咳毒素（PT）處理組(PT組，n=30)，免疫組化、westernblotting分別測(cè)定缺血再灌注后6h、12h、24h各組中海馬神經(jīng)內(nèi)Gαi2的存在及表達(dá)，流式細(xì)胞術(shù)測(cè)定各組中海馬細(xì)胞內(nèi)游離Ca~（2+）濃度的平均熒光值，分析比較它們之間的關(guān)系，TUNEL法檢測(cè)神經(jīng)元細(xì)胞凋亡。 結(jié)果相對(duì)應(yīng)時(shí)間點(diǎn)，IR組Gαi2含量和Ca~（2+）濃度較SS組明顯增高(P＜0.01)，PT組Gαi2表達(dá)量較IR組無(wú)顯著變化(P0.05)，，PT組Ca~（2+）濃度較IR組升高(P＜0.01)，PT組神經(jīng)細(xì)胞凋亡率較IR組升高(P＜0.05)。 論Gαi2在大鼠腦缺血再灌注損傷中表達(dá)明顯增高，并可降低缺血神經(jīng)細(xì)胞內(nèi)的鈣離子濃度，減少神經(jīng)細(xì)胞的凋亡，對(duì)缺血神經(jīng)細(xì)胞具有保護(hù)作用。
[Abstract]:Objective to investigate the expression of inhibitory G protein 偽 subunit G 偽 I 2 in cerebral ischemia-reperfusion injury in rats and its effect on intracellular calcium concentration and apoptosis.Methods 90 Sprague-Dawley rats were randomly divided into three groups: sham operation group (SS group), cerebral ischemia reperfusion group (IR group), cerebral ischemia reperfusion group (IR group), cerebral ischemia reperfusion group (PT group) and intraventricular pertussis toxin (PTT) treatment group (n = 30).The presence and expression of G 偽 I 2 in hippocampal nerve,Flow cytometry (FCM) was used to measure the mean fluorescence of free Ca~(2 in hippocampal cells. The relationship between them was analyzed and Tunel was used to detect the apoptosis of neurons.The expression of G 偽 I 2 in cerebral ischemia-reperfusion injury in rats was significantly increased, and the Ca 2 + concentration in ischemic nerve cells was decreased, the apoptosis of nerve cells was reduced, and the protective effect of G 偽 I 2 on ischemic nerve cells was observed.
