吞噬及饑餓誘導(dǎo)巨噬細(xì)胞自噬對(duì)其吞噬功能的影響

【摘要】:目的研究饑餓誘導(dǎo)的自噬對(duì)巨噬細(xì)胞吞噬雞紅細(xì)胞的影響,初步探討自噬在巨噬細(xì)胞吞噬功能中的作用。方法實(shí)時(shí)動(dòng)態(tài)觀察巨噬細(xì)胞吞噬雞紅細(xì)胞的過(guò)程,單丹磺酰尸胺(MDC)熒光染色標(biāo)記巨噬細(xì)胞的自噬體,激光掃描共聚焦顯微鏡下進(jìn)行觀察和定量分析。Western blot方法檢測(cè)巨噬細(xì)胞的微管相關(guān)蛋白1輕鏈3(LC3)的表達(dá),HE染色觀察自噬對(duì)其吞噬功能的影響。結(jié)果巨噬細(xì)胞活躍地吞噬雞紅細(xì)胞,吞噬后6 h MDC熒光染色陽(yáng)性率明顯高于對(duì)照組。饑餓誘導(dǎo)巨噬細(xì)胞LC3Ⅱ表達(dá)增高,并可以被3-甲基腺嘌呤(3-MA)抑制,饑餓3 h后的巨噬細(xì)胞對(duì)雞紅細(xì)胞的吞噬率與對(duì)照組相比無(wú)明顯變化。自噬被3-MA抑制后,巨噬細(xì)胞對(duì)雞紅細(xì)胞的吞噬率明顯低于對(duì)照組。結(jié)論巨噬細(xì)胞活躍地吞噬雞紅細(xì)胞,吞噬后自噬明顯增強(qiáng);自噬對(duì)巨噬細(xì)胞吞噬功能有重要作用。
[Abstract]:Objective to study the effect of starvation induced autophagy on phagocytosis of chicken erythrocytes and the role of autophagy in phagocytosis of macrophages.Methods the process of phagocytosis of chicken erythrocytes by macrophages was observed in real time.The effect of autophagy on phagocytosis was observed by HE staining and the expression of microtubule-associated protein 1 light chain 3LC-3 in macrophages was detected by Western blot.Results the positive rate of MDC fluorescence staining of macrophages was significantly higher than that of the control group 6 h after phagocytosis.The expression of LC3 鈪,
