【摘要】:目的為了評(píng)價(jià)BCG初免-單次重組腺病毒Ad5-CEAB加強(qiáng)的免疫策略誘導(dǎo)小鼠產(chǎn)生的免疫效應(yīng)。方法在前期工作的基礎(chǔ)上,制備并純化獲得了高滴度的重組腺病毒Ad5-CEAB,然后以小鼠為動(dòng)物模型,采用BCG初免-Ad5-CEAB加強(qiáng)的異源初免-加強(qiáng)的免疫策略對(duì)動(dòng)物進(jìn)行免疫,通過(guò)測(cè)定小鼠淋巴細(xì)胞抗原刺激液中IL-12和IFN-γ的含量,測(cè)定小鼠血清IgG抗體含量以及小鼠肺盥洗液中sIgA的濃度來(lái)評(píng)價(jià)BCG初免-單次重組腺病毒Ad5-CEAB加強(qiáng)免疫策略的免疫效應(yīng)。結(jié)果 BCG初免-Ad5-CEAB加強(qiáng)免疫后,經(jīng)抗原刺激后的小鼠淋巴細(xì)胞會(huì)分泌高水平的IL-12和IFN-γ,這兩種細(xì)胞因子的濃度均顯著高于對(duì)照組。BCG初免-Ad5-CEAB加強(qiáng)后,還可以有效的刺激小鼠分泌IgG抗體,血清IgG抗體濃度顯著高于對(duì)照組。另外,小鼠肺盥洗液中sIgA的濃度也顯著高于對(duì)照組。結(jié)論采用BCG初免-單次Ad5-CEAB加強(qiáng)的異源初免-加強(qiáng)的免疫策略能夠有效的刺激小鼠產(chǎn)生較強(qiáng)的免疫應(yīng)答反應(yīng)。
[Abstract]:Objective to evaluate the immune effect induced by BCG primary immune-single recombinant adenovirus Ad5-CEAB enhanced immunization strategy in mice. Methods on the basis of previous work, The recombinant adenovirus Ad5-CEAB with high titer was prepared and purified. Then mice were used as an animal model to immunize animals with the strategy of allogeneic immune-enhancement enhanced by BCG primary immune-Ad5-CEAB. The contents of IL-12 and IFN- 緯 in mouse lymphocyte antigen stimulating fluid were determined. The level of serum IgG antibody and the concentration of sIgA in mouse lung lavatory fluid were determined to evaluate the immune effect of BCG primary immune-single recombinant adenovirus Ad5-CEAB. The lymphocytes stimulated by antigen could secrete high levels of IL-12 and IFN- 緯. The concentrations of these two cytokines were significantly higher than those of the control group. The concentration of the two cytokines was significantly higher than that of the control group. After immunized with -Ad5-CEAB, they could effectively stimulate the secretion of IgG antibody in mice. The concentration of serum IgG antibody was significantly higher than that of the control group. The concentration of sIgA in mouse lung lavatory solution was also significantly higher than that in control group. Conclusion the strategy of BCG first immune-single time Ad5-CEAB enhanced xenogeneic immune-enhancement can effectively stimulate the mice to produce a strong immune response.
