【摘要】：目的通過觀察血尿膠囊對急性腎盂腎炎模型大鼠臟器指數(shù)、細(xì)胞因子以及腎臟組織的影響,探討血尿膠囊治療急性腎盂腎炎的作用機(jī)制。方法采用"半結(jié)扎"輸尿管,膀胱注射大腸桿菌方法制備急性腎盂腎炎大鼠模型,治療組于造模后第3天起每天給藥1次,正常組及模型組給予等量生理鹽水,連續(xù)10 d,第13天處死動物,檢測臟器指數(shù),測定血清中血肌酐、IL-1α(白細(xì)胞介素-1α)、IL-1β、IL-6、IL-10、CXCL-2(趨化因子-2)的含有量,對右側(cè)結(jié)扎腎臟進(jìn)行細(xì)菌培養(yǎng)并行病理組織學(xué)檢查。結(jié)果大鼠造模后,左右腎比值、腎臟指數(shù)增大(P0.01),膀胱指數(shù)有所升高(P0.05);造模大鼠IL-1α、IL-1β、IL-6、IL-10、血肌酐、尿肌酐、CXCL-2含有量均顯著升高(P0.05,P0.01);模型組大鼠右側(cè)結(jié)扎腎臟細(xì)菌培養(yǎng)陽性率為80%;給藥10 d后,血尿膠囊高、中劑量組左右腎比值、腎臟指數(shù)下降(P0.01),接近于空白組,膀胱指數(shù)變化不明顯;血尿膠囊高、中劑量組IL-1α、IL-6、IL-10含有量均明顯下降(P0.01),血尿膠囊高劑量組IL-1β含有量下降明顯(P0.01),血尿膠囊高、中、低劑量組尿肌酐、CXCL-2均有所降低(P0.05,P0.01),血肌酐含有量并無明顯變化(P0.05);血尿膠囊高劑量組細(xì)菌培養(yǎng)陽性率為20%,中劑量組為50%,低劑量組為60%;病理組織檢查結(jié)果顯示血尿膠囊能夠顯著降低急性腎盂腎炎大鼠腎臟感染、減輕炎癥反應(yīng)。結(jié)論血尿膠囊能夠改善腎功能,修復(fù)腎小球基底膜,降低尿肌酐含有量,下調(diào)趨化因子水平,減輕急性腎盂腎炎模型大鼠的腎臟炎癥反應(yīng),達(dá)到治療急性腎盂腎炎的目的。
[Abstract]:Objective to investigate the mechanism of Xueniao capsule in treating acute pyelonephritis by observing the effects of Xueniao capsule on organ index, cytokines and renal tissue in rats with acute pyelonephritis. The model of acute pyelonephritis was made by intravesical injection of Escherichia coli. The rats in the treatment group were given the same amount of normal saline once a day from the 3rd day after the model, and the normal group and the model group were given the same amount of normal saline for 10 days. The animals were killed on the 13th day and the visceral index was measured. Serum levels of serum creatinine interleukin-1 偽 (IL-1 偽) were determined, and the ratio of left and right kidneys was determined by bacterial culture and histopathological examination after the right ligation of the kidney, and the contents of CXCL-2 (chemokine 2) were measured. The positive rate of IL-1 偽 IL-1 尾 IL-6 IL-10, serum creatinine, urinary creatinine CXCL-2 was significantly increased in the model rats, and the positive rate of the right side ligation of kidney bacteria was 80 in the model group, 10 days after administration, the blood and urine capsules were high. In the middle dose group, the ratio of left and right kidney, the decrease of renal index and the decrease of renal index were close to the blank group, but the bladder index did not change obviously, the content of IL-1 偽 IL-6 IL-10 in the middle dose group decreased significantly, and the content of IL-1 尾 decreased significantly in the high dose group of Xueniao capsule, and the content of IL-1 尾 in the high dose group was significantly lower than that in the control group, and the content of IL-10 in Xueliao capsule was higher than that in the control group. Urinary creatinine CXCL-2 was decreased in low dose group, and serum creatinine content was not significantly changed. The positive rate of bacterial culture was 20 in the high dose group, 50 in the middle dose group and 60 in the low dose group. Pathological examination showed that the blood creatinine content of Xueniao capsule was 60%, and that of the high dose group was 20%, while that of the middle dose group was 50%, and that of the low dose group was 60%. Can significantly reduce renal infection in rats with acute pyelonephritis. Conclusion Xueliao capsule can improve renal function, repair glomerular basement membrane, reduce urinary creatinine content, down-regulate chemokine level, and alleviate renal inflammation in rats with acute pyelonephritis. To achieve the treatment of acute pyelonephritis.
