姜黃素對(duì)PD模型細(xì)胞活性氧簇水平及沉默信息調(diào)控因子3表達(dá)的影響

【摘要】:目的探討姜黃素對(duì)魚藤酮誘導(dǎo)的帕金森病(PD)模型SH-SY5Y細(xì)胞活性氧簇(ROS)水平及沉默信息調(diào)控因子3(Sirt3)表達(dá)的影響,為PD的臨床治療提供依據(jù)。方法將體外培養(yǎng)的SH-SY5Y細(xì)胞分為姜黃素1組、姜黃素2組、姜黃素3組、姜黃素4組、模型組和對(duì)照組。姜黃素1、2、3、4組先分別加入0.5、1.0、5.0、10.0μmol/L的姜黃素培養(yǎng)4 h,然后加入0.1μmol/L魚藤酮共同培養(yǎng)24 h;模型組直接加入0.1μmol/L魚藤酮培養(yǎng)24 h;對(duì)照組培養(yǎng)基內(nèi)不加任何藥物培養(yǎng)24 h。采用DCFH-DA染色法檢測(cè)各組ROS水平,RT-PCR法測(cè)定Sirt3 mRNA相對(duì)表達(dá)量,Western blotting法測(cè)定Sirt3蛋白相對(duì)表達(dá)量。結(jié)果 ROS水平:模型組高于對(duì)照組(P0.05),姜黃素2組姜黃素1組模型組(P均0.05),但姜黃素3組、姜黃素4組、模型組兩兩比較P均0.05。Sirt3 mRNA和蛋白相對(duì)表達(dá)量:模型組低于對(duì)照組(P均0.05),姜黃素2組姜黃素1組模型組(P均0.05),但姜黃素3組、姜黃素4組、模型組兩兩比較P均0.05。結(jié)論姜黃素可降低SH-SY5Y細(xì)胞ROS水平,上調(diào)Sirt3表達(dá);此作用姜黃素濃度為1.0μmol/L時(shí)最為明顯。
[Abstract]:Objective to investigate the effect of curcumin on the expression of rotenone-induced rotenone-induced reactive oxygen species (Ros) in SH-SY5Y cells and the expression of silencing signal regulator 3- Sirt3. Methods SH-SY5Y cells cultured in vitro were divided into three groups: curcumin group 1, curcumin group 2, curcumin group 3, curcumin group 4. The model group and the control group were cultured for 4 h respectively with 0. 5 渭 mol/L rotenone for 0. 5 渭 mol/L and then 0. 1 渭 mol/L rotenone for 24 h respectively. The model group was cultured directly with 0. 1 渭 mol/L rotenone for 24 h; the control medium was not cultured with any drugs. The relative expression of Sirt3 mRNA in each group was detected by DCFH-DA staining method and the relative expression of Sirt3 mRNA by RT-PCR. Results ROS level of model group was higher than that of control group (P 0.05), curcumin group 2 was curcumin group 1 model group. P, 0.05, but curcumin group 3, The relative expression of 0.05.Sirt3 mRNA and protein in the model group was lower than that in the control group (P < 0.05), curcumin group 1 model group (P 0.05), curcumin group 3, curcumin group 4, but curcumin group 3 and curcumin group 4. Conclusion curcumin can decrease the level of ROS and up-regulate the expression of Sirt3 in SH-SY5Y cells, especially when the concentration of curcumin is 1.0 渭 mol/L.
