生長(zhǎng)激素對(duì)人外周血來(lái)源NK細(xì)胞殺傷活性的影響及機(jī)制探討

【摘要】:目的觀察生長(zhǎng)激素(GH)對(duì)人外周血來(lái)源自然殺傷(NK)細(xì)胞殺傷活性的影響,并探討其機(jī)制。方法選取10名健康志愿者,采用免疫磁珠法分離并培養(yǎng)其外周血NK細(xì)胞。將NK細(xì)胞分為A、B、C組,分別與終濃度0、5、20μg/mL的重組人GH共培養(yǎng)48 h。以人類髓性白血病K562細(xì)胞為靶細(xì)胞,NK細(xì)胞為效應(yīng)細(xì)胞,按不同的效靶比(25∶1、50∶1、100∶1)混合,采用CCK-8法檢測(cè)NK細(xì)胞對(duì)K562細(xì)胞的殺傷率;取各組培養(yǎng)48 h的NK細(xì)胞,分別采用流式細(xì)胞術(shù)和Real-time RT-PCR法檢測(cè)NK細(xì)胞表面活化受體NKG2D蛋白及基因。結(jié)果三個(gè)不同效靶比下,B、C組NK細(xì)胞對(duì)K562細(xì)胞的殺傷率高于A組,且C組高于B組(P均0.01)。A、B、C組NKG2D陽(yáng)性表達(dá)的NK細(xì)胞百分?jǐn)?shù)分別為78.8%±2.8%、87.5%±3.1%、91.8%±3.3%,B、C組NKG2D陽(yáng)性表達(dá)的NK細(xì)胞百分?jǐn)?shù)高于A組(P均0.05)。A、B、C組NKG2D mRNA相對(duì)表達(dá)量分別為1±0.01、5±2.32、10±3.21,B、C組NKG2D mRNA相對(duì)表達(dá)量高于A組,且C組高于B組(P均0.05)。結(jié)論 GH可通過(guò)上調(diào)NK細(xì)胞NKG2D的表達(dá),增強(qiáng)其殺傷活性。
[Abstract]:Objective to observe the effect of growth hormone (GH) on the cytotoxicity of NK cells from human peripheral blood and its mechanism. Methods Ten healthy volunteers were selected. NK cells in peripheral blood were isolated and cultured by immunomagnetic bead method. NK cells were divided into two groups: group C, co-cultured with recombinant human GH with final concentration of 0 ~ 5 渭 g/mL for 48 h. Human myeloid leukemia K562 cells were used as target cells and NK cells were used as effector cells. The cytotoxicity of NK cells to K562 cells was detected by CCK-8 assay, and the NK cells were cultured for 48 h. Flow cytometry and Real-time RT-PCR were used to detect the surface activated receptor (NKG2D) protein and gene of NK cells. Results the cytotoxicity of NK cells to K562 cells was higher in group C than in group A. The percentage of NKG2D positive NK cells in group C was higher than that in group B (78.8% 鹵2.8% 鹵3.1% 鹵3.1% 鹵3.1% 鹵3.31%), respectively. The relative expression of NKG2D mRNA in group C was higher than that in group A (1 鹵0.01 鹵2.32 10 鹵3.21). The relative expression of NKG2D mRNA in group C was higher than that in group A (P = 0.05 鹵2.32 10 鹵3.21), and the percentage of NK cells in group C was higher than that in group A (P = 0.05 鹵2.32 10 鹵3.21), and the percentage of NK cells in group C was higher than that in group A (P 0.05 鹵2.32 10 鹵3.21). Conclusion GH can enhance the cytotoxicity of NK cells by upregulating the expression of NKG2D.
