人羊膜臍帶源性間充質(zhì)干細(xì)胞對(duì)大鼠肝硬化的治療作用

【摘要】:目的:比較人臍帶間充質(zhì)干細(xì)胞(human amniotic membrane-derived mesenchymal stem cells,hAM-MSCs)和人羊膜間充質(zhì)干細(xì)胞(human umbilical cord-derived mesenchymal stem cells,hUC-MSCs)對(duì)模型大鼠肝硬化的治療效果.方法:分離培養(yǎng)hAM-MSCs和hUC-MSCs,流式檢測(cè)CD29、CD44和CD34.CCl4誘導(dǎo)大鼠肝硬化模型,在第8周時(shí),殺死5只大鼠做病理檢測(cè)以確診為肝硬化.30只成模大鼠隨機(jī)分為臍帶干細(xì)胞組(n=10)、羊膜干細(xì)胞組(n=10)和對(duì)照組(n=10),分別注入hAM-MSCs、hUC-MSCs2mL(細(xì)胞數(shù)量2×106)和等量生理鹽水.細(xì)胞移植前和移植4wk后,檢測(cè)大鼠肝功能;肝組織HE染色、Masson染色;免疫組織化學(xué)法檢測(cè)α-SMA在肝臟中的表達(dá).結(jié)果:2種細(xì)胞表達(dá)CD29、CD44,不表達(dá)CD34.移植后,與對(duì)照組相比,臍帶干細(xì)胞組和羊膜干細(xì)胞組ALT和AST明顯降低(204.6±16.4,195.6±21.2vs539.8±36.2;180.1±25.2,167.5±19.0vs337.4±23.4,P0.05);膠原沉積減少;組織病理學(xué)評(píng)分有顯著性差異(P0.05);肝臟α-SMA表達(dá)量減少(130.6±3.0,127.0±2.6vs152.2±5.4,P0.05).臍帶干細(xì)胞組和羊膜干細(xì)胞組的肝功能、肝組織病理學(xué)評(píng)分及肝臟α-SMA表達(dá)量均無明顯差異.結(jié)論:hAM-MSCs和hUC-MSCs能有效改善肝硬化大鼠模型的肝功能和肝硬化程度,兩者治療效果無明顯差異.
[Abstract]:Objective: to compare the therapeutic effects of human amniotic membrane-derived mesenchymal stem cells (hAM-MSCs) and human umbilical cord-derived mesenchymal stem cells (hUC-MSCs) on liver cirrhosis in model rats. Methods: hAM-MSCs and hUC-MSCswere isolated and cultured, and CD29CD44 and CD34.CCl4 induced rat models of cirrhosis were detected by flow cytometry. At week 8, Five rats were killed to make pathological examination to confirm cirrhosis. 30 model rats were randomly divided into umbilical cord stem cell group and amniotic membrane stem cell group, and control group were injected with hAM-MSCshUC-MSCs 2mL (number of cells 2 脳 106) and normal saline respectively. Before and 4 wk after transplantation, the cells were injected with hAM-MSCsHUC-MSCs 2mL (number of cells 2 脳 106) and the same amount of normal saline respectively. The expression of 偽 -SMA in liver was detected by immunohistochemical method. Results two kinds of cells expressed CD29 CD44, not CD34. After transplantation, the expression of 偽 -SMA was compared with that of control group. In cord cord stem cell group and amniotic membrane stem cell group, ALT and AST decreased significantly (195.6 鹵21.2vs539.8 鹵36.2n 180.1 鹵25.2vs337.4 鹵23.4P0.05; collagen deposition decreased; histopathological score was significantly different P0.05; liver 偽 -SMA expression decreased 130.6 鹵3.0V 127.0 鹵2.6vs152.2 鹵5.4P0.05. the liver function of cord cord stem cell group and amniotic stem cell group was significantly lower than that of amniotic stem cell group. There was no significant difference in liver histopathological score and 偽 -SMA expression in liver. Conclusion the liver function and the degree of liver cirrhosis in rats with liver cirrhosis can be effectively improved by hUC-MSCs and hAM-MSCs, and there is no significant difference in the therapeutic effect between the two groups.
