銀杏葉提取物(EGb761)對(duì)人脂肪間充質(zhì)干細(xì)胞神經(jīng)向分化的影響

【摘要】:目的:觀察銀杏葉提取物(EGb761)對(duì)人脂肪間充質(zhì)干細(xì)胞(hADSCs)增殖及神經(jīng)向分化的影響。 方法:用CCK-8法檢測(cè)不同濃度EGb761(40mg/L、80mg/L、120mg/L)作用下hADSCs的增殖情況;含有不同濃度EGb761(40mg/L、80mg/L、120mg/L)的聯(lián)合神經(jīng)誘導(dǎo)因子(20ng/ml bFGF、20ng/ml EGF、20ng/ml BDNF)對(duì)hADSCs神經(jīng)向誘導(dǎo)分化,觀察形態(tài)學(xué)變化,免疫熒光檢測(cè)Nestin、NSE、GFAP于3d、7d、10d、14d的表達(dá)情況,計(jì)數(shù)陽(yáng)性細(xì)胞率。 結(jié)果:EGb761對(duì)hADSCs增殖的影響具有時(shí)間差異性,1d時(shí)無(wú)明顯差異,3d時(shí)顯著促進(jìn),5d時(shí)促增殖趨于平緩,7d時(shí)細(xì)胞有增殖,但增殖幅度減小;含有不同濃度EGb761的聯(lián)合誘導(dǎo)因子促進(jìn)hADSCs的形態(tài)學(xué)發(fā)生神經(jīng)向分化,與陽(yáng)性對(duì)照組相比各時(shí)間點(diǎn)均促進(jìn)Nestin的表達(dá),,且有濃度依賴趨勢(shì);促進(jìn)NSE的表達(dá),無(wú)明顯濃度依賴;GFAP的表達(dá)與陽(yáng)性對(duì)照無(wú)差異。 結(jié)論:EGb761對(duì)hADSCs的增殖有促進(jìn)作用,3d時(shí)促進(jìn)作用明顯;EGb761顯著促進(jìn)hADSCs的神經(jīng)向分化,不促進(jìn)向膠質(zhì)樣細(xì)胞分化。
[Abstract]:Aim: to observe the effects of Ginkgo biloba extract (EGb761) on the proliferation and neural differentiation of human adipose mesenchymal stem cells (hADSCs). Methods: CCK-8 method was used to detect the proliferation of hADSCs induced by different concentrations of EGb761mg / L 80mg / L (80mg / L), and the expression of Nestin NSEGFAP was detected at 3d7 d 10d ~ 14d by combined nerve inducible factor 20ng / ml bFGF20ngr / ml EGFGF20ngr / ml. The expression of Nestin NSEGFAP was detected by immunofluorescence. The positive cell rate was counted. Results the effects of 1% EGb761 on the proliferation of hADSCs showed no significant difference at 1 day. At 3 days, the proliferation of hADSCs increased significantly at 5 days and tended to be smooth at 7 days, but the range of proliferation decreased. The co-induction factors containing different concentrations of EGb761 promoted the morphological differentiation of hADSCs, and all of them promoted the expression of Nestin in a concentration-dependent manner, and promoted the expression of NSE in a dose-dependent manner, compared with the positive control group. There was no significant difference in the expression of GFAP between the two groups. ConclusionEGB 761 can promote the proliferation of hADSCs. EGb761 can obviously promote the differentiation of hADSCs nerve, but not to glial cells.
