【摘要】：目的觀察注意力缺陷多動障礙(attention deficit hyperactivity disorder,ADHD)模型大鼠注意定勢轉(zhuǎn)移能力變化及靜寧顆粒對其的影響。方法采用自發(fā)性高血壓大鼠(spontaneously hypertensive rats,SHR)作為ADHD的模型大鼠,分別給予靜寧顆粒(20.25、10.125、5.062 5 g/kg)、鹽酸哌甲酯緩釋片(利他林6.75 mg/kg)和小兒黃龍顆粒(0.937 5 g/kg),連續(xù)給藥17 d進行治療。以大鼠注意定勢轉(zhuǎn)移任務(wù)(Attentional set-shifting,AST)中錯誤次數(shù)、挖掘次數(shù)、錯誤率等行為學(xué)指標(biāo),評價藥物的藥效。結(jié)果與正常對照組比較,模型大鼠在簡單辨別(SD)階段、復(fù)雜辨別(CD)階段、內(nèi)維度轉(zhuǎn)換(IDS)階段、逆反學(xué)習(xí)(RL)階段、外維度轉(zhuǎn)換(EDS)階段錯誤次數(shù)、錯誤率、挖掘次數(shù)均增加;與模型組比較,靜寧顆粒連續(xù)給藥17 d后,各劑量可降低模型大鼠錯誤次數(shù)、錯誤率、挖掘次數(shù),改善ADHD模型大鼠注意定勢轉(zhuǎn)移缺陷。結(jié)論 ADHD模型大鼠具有注意定勢轉(zhuǎn)移缺陷,靜寧顆�？捎行Ц纳艫DHD模型大鼠的注意定勢轉(zhuǎn)移能力。
[Abstract]:Objective to observe the change of attention setting transfer ability of attention deficit hyperactivity disorder (ADHD) rats and the effect of Jingning granule on it. Methods spontaneous hypertensive rats were used as model rats of ADHD. Jingning granules were given 20. 25 渭 g / kg 10. 125n 5. 062 5 g / kg, 6. 75 mg / kg Ritalin hydrochloride sustained release tablets and 0. 937 5 g / kg Huanglong granule for 17 days. The number of errors and the number of excavations in the attention set transfer task of rats were observed, and the number of times of excavating the drug was compared with that of Ritalin hydrochloride sustained release tablet (Ritalin 6. 75 mg / kg) and Huanglong granule of children (0. 937 5 g / kg) for 17 days. Results compared with the normal control group, the model rats were in the simple discrimination stage, the complex discrimination stage, the internal dimension conversion stage and the inverse learning stage. The number of errors, the error rate and the times of mining were increased in the outer dimension conversion EDS stage, and compared with the model group, after 17 days of continuous administration of Jingning granule, the number of errors, the error rate and the times of mining were decreased in each dose of Jingning granule. Conclusion ADHD model rats have attentional homeostasis defect and Jingning granule can effectively improve the attention set transfer ability of ADHD model rats.
