良性前列腺增生合并遲發(fā)性性腺功能減退大鼠模型的構(gòu)建

【摘要】:目的探討良性前列腺增生(BPH)合并遲發(fā)性性腺功能減退癥(LOH)動物模型的建立方法,并分析其特點。方法選取80只SD大鼠,隨機分為對照組、BPH合并LOH組、BPH組、LOH組,每組均為20只。BPH合并LOH組給予環(huán)磷酰胺[20 mg/(kg·d)]腹腔內(nèi)連續(xù)注射5d,而后給予丙酸睪酮[50 mg/(kg·d)]腹腔內(nèi)連續(xù)注射28d;BPH組以同等體積生理鹽水代替環(huán)磷酰胺,LOH組以同等體積橄欖油代替丙酸睪酮,對照組以同等體積生理鹽水代替環(huán)磷酰胺、同等體積橄欖油代替丙酸睪酮,給藥方法同BPH合并LOH組。停藥2d后,進(jìn)行懸尾實驗、負(fù)重游泳實驗及交配實驗,并通過斷尾法取血以檢測血清睪酮含量。完成全部操作后,通過頸椎脫臼法處死全部實驗動物,解剖觀察前列腺、稱量并行組織病理學(xué)檢查。結(jié)果 LOH組、BPH合并LOH組、對照組、BPH組大鼠平均前列腺指數(shù)分別為1.58±0.13、2.93±0.19、2.33±0.13、3.23±0.11,血清睪酮含量分別為(4.91±1.06)、(9.52±1.02)、(12.59±0.70)、(19.69±0.56)ng/mL,懸尾掙扎次數(shù)分別為(97.40±15.86)、(120.40±14.06)、(223.83±16.51)、(235.29±18.77)次,各組間差異均有統(tǒng)計學(xué)意義(P0.05,P0.01);LOH組、BPH合并LOH組、對照組、BPH組大鼠平均負(fù)重游泳時間分別為(74.27±9.29)、(167.47±23.35)、(302.33±30.10)、(261.59±35.13)s,嗅陰次數(shù)分別為(1.53±0.52)、(3.07±0.88)、(9.17±1.30)、(9.59±1.12)次,除對照組和BPH組差異無統(tǒng)計學(xué)意義外,其余各組間差異均有統(tǒng)計學(xué)意義(P0.01);LOH組、BPH合并LOH組、對照組、BPH組大鼠平均騎跨次數(shù)分別為(0.33±0.49)、(0.47±0.52)、(2.11±0.47)、(2.29±0.47)次,除對照組與BPH組、BPH合并LOH組與LOH組差異無統(tǒng)計學(xué)意義外,其余各組間差異均有統(tǒng)計學(xué)意義(P0.01)。結(jié)論通過腹腔注射環(huán)磷酰胺及丙酸睪酮可獲得BPH合并LOH大鼠模型。
[Abstract]:Objective to investigate the method of establishing the animal model of benign prostatic hyperplasia (BPH) with delayed hypogonadism and to analyze its characteristics. Methods Eighty SD rats were randomly divided into two groups: the control group (BPH + LOH group) and the control group (LOH group). 20 rats in each group were given cyclophosphamide [20 mg/(kg 路d] intraperitoneal injection for 5 days, and then testosterone propionate [50 mg/(kg 路d] was injected intraperitoneally for 28 days. The same volume of normal saline was used instead of cyclophosphamide loh group. Oil instead of testosterone propionate, The control group was given the same volume of normal saline instead of cyclophosphamide, the same volume of olive oil instead of testosterone propionate, and the same dosage of BPH combined with LOH group. After 2 days, suspension test, weight-bearing swimming test and mating experiment were carried out. After the complete operation, all the experimental animals were killed by cervical dislocations, the prostate was dissected, the prostate was measured and histopathological examination was performed. Results the LOH group was treated with LOH. The average prostatic index of rats in the control group was 1.58 鹵0.131.93 鹵0.19 鹵2.33 鹵0.133.23 鹵0.11, and the serum testosterone level was 4.91 鹵1.06 鹵1.06 鹵1.06 鹵12.59 鹵0.70 鹵19.69 鹵0.56 ng / mL, respectively. The number of tail struggle was 97.40 鹵15.86 ng / mL. 40 鹵14.063.83 鹵16.51D 235.29 鹵18.77). There were significant differences between the two groups. The average weight-bearing swimming time of rats in the control group was 74.27 鹵9.29, 167.47 鹵23.35, 302.33 鹵30.10, 261.59 鹵35.13 s, and the number of olfactory negative was 1.53 鹵0.52, 3.07 鹵0.882.17 鹵1.309.59 鹵1.12, respectively. Except for the control group and the BPH group, the differences were statistically significant. The average spanking times of rats in the control group were 0.33 鹵0.49, 0.47 鹵0.52, 2.11 鹵0.47, 2.29 鹵0.47) respectively, except that there was no significant difference between the control group and the BPH group in the combination of LOH group and LOH group. Conclusion BPH combined with LOH rat model can be obtained by intraperitoneal injection of cyclophosphamide and testosterone propionate.
