紋狀體神經(jīng)元調(diào)控SVZ區(qū)域神經(jīng)再生的作用機(jī)制

【摘要】:目的:紋狀體神經(jīng)元可以投射突觸到室管膜下區(qū)(SVZ),并影響SVZ區(qū)域前體細(xì)胞的活性,但是紋狀體神經(jīng)元活性如何影響SVZ區(qū)域神經(jīng)再生未知,本文旨在探討紋狀體神經(jīng)元與SVZ區(qū)域神經(jīng)再生的關(guān)系。方法:利用光遺傳學(xué)的方法,特異調(diào)控紋狀體神經(jīng)元活性后,利用免疫組織化學(xué)方法觀察其活性對(duì)SVZ區(qū)域神經(jīng)再生的影響,并且探討其可能機(jī)制。結(jié)果:當(dāng)以CaM KⅡ作為啟動(dòng)子時(shí),光敏感蛋白可以在紋狀體神經(jīng)元中表達(dá),而且紋狀體神經(jīng)元的發(fā)放頻率能夠被激光控制。紋狀體神經(jīng)元活性的抑制促進(jìn)NT-1的分泌(P0.05),免疫組織化學(xué)結(jié)果顯示紋狀體神經(jīng)元活性的降低促進(jìn)SVZ區(qū)域BrdU與DCX雙陽(yáng)性細(xì)胞數(shù)目的增多(P0.05)。結(jié)論:紋狀體神經(jīng)元活性是影響SVZ區(qū)域神經(jīng)再生的重要因素,抑制紋狀體神經(jīng)元活性可以促進(jìn)神經(jīng)再生。
[Abstract]:Objective: striatal neurons can project synapses into the subependymal area of Sv Z, and affect the activity of precursor cells in the SVZ region. However, how the activity of striatal neurons affects the regeneration of neurons in the SVZ region is unknown. The aim of this study was to investigate the relationship between striatal neurons and the regeneration of neurons in the SVZ region.
