弓形蟲ROP18蛋白對(duì)宿主細(xì)胞凋亡的影響

【摘要】:目的:研究弓形蟲棒狀體蛋白R(shí)OP18是否與弓形蟲抑制宿主細(xì)胞的凋亡有關(guān)。 方法:利用軟件設(shè)計(jì)擴(kuò)增ROP18基因的引物,在ROP18基因上游引入Flag標(biāo)簽序列,為方便下游克隆,在上游及下游引物的5’端分別引入BamHⅠ及Hind Ⅲ酶切位點(diǎn)。酚-氯仿法提取弓形蟲RH株基因組DNA,,以其為模板PCR擴(kuò)增ROP18基因。利用試劑盒純化PCR產(chǎn)物,利用BamHⅠ和Hind Ⅲ雙酶切克隆到pcDNA3.1(+)真核表達(dá)載體的相同位點(diǎn),構(gòu)建pcDNA3.1(+)-Flag-ROP18真核表達(dá)載體,DNA序列分析鑒定正確后,大量提取質(zhì)粒,利用試劑盒去除內(nèi)毒素,脂質(zhì)體法轉(zhuǎn)染RAW264.7細(xì)胞表達(dá)。分離蛋白,SDS-PAGE及利用Flag標(biāo)簽抗體進(jìn)行Western blotting檢測(cè)ROP18在RAW264.7細(xì)胞中的表達(dá)。H2O2分別誘導(dǎo)不含重組質(zhì)粒的RAW264.7細(xì)胞(對(duì)照組)、含重組質(zhì)粒的RAW264.7細(xì)胞(轉(zhuǎn)染組)發(fā)生凋亡,于24h、48h分別檢測(cè)以下凋亡指標(biāo):細(xì)胞爬片經(jīng)HE染色后顯微鏡下觀察細(xì)胞形態(tài)的變化,觀察細(xì)胞是否表現(xiàn)凋亡的特征性形態(tài)變化;分離線粒體及細(xì)胞漿進(jìn)行Western blotting檢測(cè)細(xì)胞色素c在線粒體與胞漿中的分布,確定細(xì)胞色素C是否從線粒體釋放到胞漿;提取細(xì)胞基因組DNA進(jìn)行瓊脂糖電泳EB染色后檢測(cè)DNA是否發(fā)生了凋亡特征性的梯狀片斷化。綜合以上實(shí)驗(yàn)結(jié)果判斷弓形蟲ROP18蛋白對(duì)H2O2誘導(dǎo)的RAW264.7細(xì)胞凋亡是否具有抑制作用。 結(jié)果: DNA序列分析結(jié)果顯示正確構(gòu)建了pcDNA3.1(+)-Flag-ROP18真核表達(dá)載體,Western blotting檢測(cè)到ROP18在RAW264.7細(xì)胞中的表達(dá)。H2O2誘導(dǎo)轉(zhuǎn)染組和對(duì)照組RAW264.7細(xì)胞凋亡的結(jié)果顯示:轉(zhuǎn)染組與對(duì)照組RAW264.7細(xì)胞形態(tài)均出現(xiàn)明顯的凋亡特征:細(xì)胞變圓,細(xì)胞膜皺縮,染色質(zhì)濃縮;Western blotting檢測(cè)到細(xì)胞色素c在線粒體與胞漿中均有分布,,說(shuō)明細(xì)胞色素c從線粒體轉(zhuǎn)移至胞漿;DNA出現(xiàn)片斷化,可見典型的DNA“梯狀”條帶,表現(xiàn)均表現(xiàn)凋亡特點(diǎn)。 結(jié)論: 1.成功構(gòu)建真核表達(dá)質(zhì)粒pcDNA3.1(+)-Flag-ROP18,該質(zhì)粒在RAW264.7細(xì)胞中表達(dá)了微量的ROP18蛋白。 2.本實(shí)驗(yàn)中表達(dá)的弓形蟲ROP18對(duì)H2O2誘導(dǎo)的RAW264.7宿主細(xì)胞凋亡無(wú)抑制作用。
[Abstract]:Aim: to investigate whether ROP18 of Toxoplasma gondii coryloid protein is associated with Toxoplasma gondii inhibiting apoptosis of host cells. Methods: the primers were designed to amplify the ROP18 gene by software, and the Flag tag sequence was introduced into the upstream of the ROP18 gene to facilitate the downstream cloning. The genomic DNAs of Toxoplasma gondii RH strain were extracted by phenol-chloroform method at the 5'end of the upstream and downstream primer, respectively. The ROP18 gene was amplified by PCR.
