條件恐懼刺激對(duì)成年大鼠杏仁核干細(xì)胞增殖及分化為神經(jīng)元的影響

【摘要】:目的:檢測(cè)條件恐懼刺激后大鼠杏仁核區(qū)神經(jīng)干細(xì)胞的數(shù)量及神經(jīng)干細(xì)胞分化為神經(jīng)元的量的變化,探討條件恐懼刺激后大鼠杏仁核區(qū)神經(jīng)干細(xì)胞的增殖及分化與恐懼記憶的關(guān)系。 方法:1.應(yīng)用足底電擊配對(duì)單調(diào)聲刺激的方法建立大鼠條件恐懼記憶模型,并以呆立時(shí)間(freezing time)評(píng)價(jià)大鼠對(duì)條件恐懼刺激事件的記憶。2.應(yīng)用免疫組化方法檢測(cè)條件恐懼刺激后不同時(shí)間點(diǎn)(1、7、14、21、30、60天)大鼠杏仁核區(qū)BrdU(外源性神經(jīng)干細(xì)胞標(biāo)記物)陽性細(xì)胞的表達(dá)量。3.應(yīng)用免疫熒光雙標(biāo)記法檢測(cè)條件恐懼刺激后不同時(shí)間點(diǎn)大鼠杏仁核區(qū)BrdU與NeuN(神經(jīng)元標(biāo)記物)雙標(biāo)陽性細(xì)胞的量。4.應(yīng)用SPSS15.0統(tǒng)計(jì)軟件進(jìn)行統(tǒng)計(jì)學(xué)分析。 結(jié)果:1.行為學(xué)檢測(cè)結(jié)果顯示:1、7、14、21、30、60天時(shí)間點(diǎn)的實(shí)驗(yàn)組大鼠在被重新放入電擊箱給予單調(diào)聲刺激后,八分鐘內(nèi)的呆立時(shí)間明顯長(zhǎng)于相應(yīng)時(shí)間點(diǎn)的對(duì)照組,差異具有顯著性(P0.01)Do。2. BrdU的免疫組織化學(xué)結(jié)果顯示:在大鼠杏仁核區(qū)可見典型的BrdU陽性細(xì)胞,計(jì)數(shù)杏仁核區(qū)BrdU陽性細(xì)胞,發(fā)現(xiàn)條件恐懼刺激后各時(shí)間點(diǎn)實(shí)驗(yàn)組大鼠杏仁核區(qū)BrdU免疫陽性細(xì)胞數(shù)少于相應(yīng)對(duì)照組,其中1、7、14、21、30天差異具有顯著性(P0.05),60天實(shí)驗(yàn)組與對(duì)照組的差異沒有統(tǒng)計(jì)學(xué)意義。3.BrdU與NeuN的免疫熒光雙標(biāo)結(jié)果顯示:在大鼠杏仁核區(qū)可見明顯的BrdU與NeuN雙標(biāo)陽性細(xì)胞,計(jì)數(shù)杏仁核區(qū)BrdU陽性細(xì)胞及BrdU與NeuN的雙標(biāo)陽性細(xì)胞,發(fā)現(xiàn)條件恐懼刺激后各時(shí)間點(diǎn)實(shí)驗(yàn)組大鼠杏仁核區(qū)BrdU和NeuN免疫熒光雙標(biāo)陽性細(xì)胞數(shù)與BrdU陽性細(xì)胞總數(shù)的比值小于相應(yīng)對(duì)照組,差異具有顯著性(P0.05)。 結(jié)論:條件恐懼刺激后大鼠呆立時(shí)間明顯延長(zhǎng),且杏仁核區(qū)細(xì)胞增殖減少。以上結(jié)果提示條件恐懼刺激有可能影響大鼠杏仁核區(qū)的神經(jīng)元新生。
[Abstract]:Objective: to detect the number of neural stem cells (NSCs) in amygdala and the number of neural stem cells (NSCs) differentiating into neurons after conditioned fear stimulation. To investigate the relationship between fear memory and proliferation and differentiation of neural stem cells in amygdala region after conditioned fear stimulation. Methods 1. The conditioned fear memory model of rats was established by plantar shock matching monotone sound stimulation. The memory of conditioned fear stimulation events in rats was evaluated by standing time and freezing time. 2. Immunohistochemical method was used to detect BrdU (exogenous neural stem cell marker) positive in amygdaloid nucleus of rats at different time points after conditioned fear stimulation. The expression of sex cells was measured by immunofluorescence double labeling method. The amount of BrdU and Neun (neuron marker) double labeled positive cells in amygdaloid nucleus of rats at different time points after conditioned fear stimulation was detected. 4. SPSS15.0 statistical software was used for statistical analysis. Results: 1.Behavioral tests showed that the rats in the experimental group at 30 days after being put back into the electric shock box for monotone sound stimulation were significantly longer in eight minutes than those in the control group at the corresponding time point. The immunohistochemical results of BrdU showed that typical BrdU positive cells were found in the amygdala of rats, and the BrdU positive cells in the amygdala were counted. It was found that the number of BrdU immunoreactive cells in the amygdala of the experimental group was less than that of the corresponding control group at each time point after conditioned fear stimulation. There was no significant difference between the experimental group and the control group on the 30th day. 3. The immunofluorescence double labeling results of BrdU and NeuN showed that there were obvious double labeled BrdU and NeuN positive cells in the amygdala of rats. BrdU positive cells and double labeled positive cells of BrdU and NeuN in amygdala were counted. It was found that the ratio of the number of BrdU and NeuN double labeled positive cells to the total number of BrdU positive cells in the amygdaloid nucleus of the experimental group was lower than that of the corresponding control group at each time point after conditioned fear stimulation, and the difference was significant (P 0.05). Conclusion: the experimental results suggest that conditioned fear stimulation may affect the neuronal growth of amygdaloid area in rats.
