芍藥內(nèi)酯苷及芍藥苷對慢性束縛應激配合孤養(yǎng)肝郁模型大鼠行為學及下丘腦-垂體-腎上腺軸的影響

【摘要】:目的:觀察芍藥內(nèi)酯苷(AF)、芍藥苷(PF)對慢性束縛應激(CRS)配合孤養(yǎng)肝郁模型大鼠行為及下丘腦-垂體-腎上腺(HPA)軸的影響,探討白芍柔肝解郁作用機制及物質(zhì)基礎。方法:將SD雄性大鼠,根據(jù)蔗糖水消耗量及體質(zhì)量隨機分組,每組12只。除空白組外,其余均采用CRS配合孤養(yǎng)復制肝郁模型。觀察體質(zhì)量,行為學指標,并用放射免疫法(RIA)檢測大鼠下丘腦促腎上腺皮質(zhì)激素釋放素(CRH)、血漿促腎上腺皮質(zhì)激素(ACTH)、皮質(zhì)酮(CORT)的含量。結果:造模第14天,與模型組比較,AF 30mg/kg組、PF 30mg/kg組大鼠體質(zhì)量明顯增加(P0.05,P0.01)。造模第21天,與模型組比較,AF 30mg/kg組、PF 30mg/kg組大鼠體質(zhì)量,曠場實驗中穿行次數(shù)明顯增加(P0.05,P0.01)而滯留時間顯著減少(P0.01),高架十字迷宮實驗OE和OT顯著增加(P0.05,P0.01),而CT顯著減少(P0.01)。AF 30mg/kg組、PF 30mg/kg組大鼠下丘腦CRH、血漿ACTH、CORT的含量均顯著降低(P0.05)。結論:AF和PF作為白芍的主要有效成分,不僅可在一定程度上改善了CRS肝郁模型大鼠的行為表現(xiàn),而且使相關應激激素的含量變化呈現(xiàn)不同的趨勢,提示二者均為白芍柔肝解郁作用的有效成分,其作用機制與抑制CRS肝郁模型大鼠HPA軸的激活有關。
[Abstract]:Objective: to observe the effects of Paeoniflorin (AFG) and Paeoniflorin (PFF) on the behavior of chronic restraint stress (CRS) and the hypothalamus-pituitary-adrenal (HPA) axis in rats with solitary liver depression. Methods: SD male rats were randomly divided into 12 rats in each group according to sucrose water consumption and body mass. The rest were treated with CRS combined with solitary nourishment to make liver depression model. Body mass and behavioral indexes were observed. The contents of corticotropin releasing hormone (CRH), plasma adrenocorticotropic hormone (ACTH) and corticosterone (Cort) in the hypothalamus of rats were determined by radioimmunoassay (RIA). Compared with the model group, the body weight of rats in the AF 30 mg / kg group increased significantly in PF 30 mg / kg group, and the body weight of the PF 30 mg / kg group was significantly increased on the 21st day of model making and the PF 30 mg / kg group compared with the model group. In open field experiment, the number of cross passage was significantly increased (P 0.05 mg / kg), and the retention time was significantly decreased. In elevated cross labyrinth test, OE and OT increased significantly (P 0.05 P 0.01), while CT significantly decreased the content of hypothalamus CRH and plasma ACTHCORT in PF 30 mg / kg group (P 0.01 路AF 30 mg / kg). Conclusion the main effective components of Radix Paeoniae Alba are: AF and PF. It can not only improve the behavior of rats with CRS liver depression to a certain extent, but also make the changes of related stress hormones present a different trend, which suggests that both of them are effective components of the effect of Radix Paeoniae Paeoniae Alba on relieving liver depression. The mechanism is related to the inhibition of activation of HPA axis in CRS liver depression model rats.
