神經(jīng)營養(yǎng)因子NT4及其受體在大鼠肺纖維化中的表達(dá)

【摘要】:實(shí)驗(yàn)?zāi)康?本研究通過對(duì)大鼠實(shí)驗(yàn)性肺纖維化中NT4及其受體TrkB的檢測(cè),來探討NT4及TrkB與大鼠肺纖維化發(fā)生發(fā)展的關(guān)系,從而研究神經(jīng)營養(yǎng)因子在肺纖維化中的變化,為肺纖維化發(fā)病機(jī)制的研究開辟新的途徑。 實(shí)驗(yàn)方法 選wistar健康雄性大鼠30只作為研究的對(duì)象,分為對(duì)照組(向氣管內(nèi)注射生理鹽水)、模型組(向氣管內(nèi)灌注博來霉素)兩組;兩組大鼠分別于造模后第7天、第14天、第28天各處死5只,取出大鼠肺組織,用Masson染色及HE染色來比較造模程度,用PCR和免疫組化檢測(cè)模型組及對(duì)照組中NT4及TrkB的含量。應(yīng)用SPSS17.0統(tǒng)計(jì)軟件進(jìn)行數(shù)據(jù)統(tǒng)計(jì)分析,單個(gè)因素的各時(shí)間點(diǎn)之間的比較,采用單因素方差分析,兩兩比較用t檢驗(yàn),并進(jìn)行相關(guān)性分析。 實(shí)驗(yàn)結(jié)果 模型組造模后第7天、第14天、第28天NT4及TrkB均有不同程度的增高,且均隨病變嚴(yán)重程度的增高而增高,結(jié)果符合統(tǒng)計(jì)學(xué)意義(P0.05),二者呈正相關(guān)(P0.001);對(duì)照組在對(duì)應(yīng)時(shí)間段二者無明顯變化(P0.05)。 實(shí)驗(yàn)結(jié)論 神經(jīng)營養(yǎng)因子NT4及其受體TrkB在肺纖維化大鼠中表達(dá)增加,可能與肺上皮細(xì)胞增生及TrkB/NT4(?)由的信號(hào)傳導(dǎo)被擾亂等因素有關(guān),說明神經(jīng)營養(yǎng)因子NT4及其受體參與肺纖維化的發(fā)生發(fā)展過程,為肺纖維化的發(fā)生提供新的論點(diǎn)。
[Abstract]:Experimental purpose The purpose of this study was to investigate the relationship between NT4 and TrkB and the occurrence and development of pulmonary fibrosis in rats by the detection of NT4 and its receptor TrkB in experimental pulmonary fibrosis. Therefore, to study the changes of neurotrophic factors in pulmonary fibrosis, and to open up a new way for the study of the pathogenesis of pulmonary fibrosis. Experimental method Thirty male wistar rats were divided into two groups: control group (intratracheal injection of normal saline) and model group (intratracheal instillation of bleomycin). Five rats were killed in each group on the 7th, 14th and 28th day after modeling. The lung tissue was taken out, and the degree of the model was compared by Masson staining and HE staining. The contents of NT4 and TrkB in model group and control group were detected by PCR and immunohistochemistry. The statistical data were analyzed by SPSS17.0 software and the time points of single factor were compared. Single factor analysis of variance (ANOVA) was used, and t test was used for pairwise comparison, and correlation analysis was carried out. Experimental results In the model group, NT4 and TrkB increased in varying degrees on the 7th, 14th and 28th day after the establishment of the model, and both increased with the severity of the lesion. The results were in accordance with the statistical significance (P 0.05), and there was a positive correlation between them. In the control group, there was no significant change in the two groups in the corresponding time period (P 0.05). Experimental conclusion The expression of neurotrophic factor NT4 and its receptor TrkB is increased in pulmonary fibrosis rats, which may be related to the proliferation of pulmonary epithelial cells and TrkB / NT4? The signal transduction was disturbed, which indicated that neurotrophic factor NT4 and its receptor were involved in the development of pulmonary fibrosis, and provided a new argument for the occurrence of pulmonary fibrosis.
