植入早期小鼠子宮內(nèi)膜形態(tài)學(xué)及Galectin-3、整合素α4表達(dá)的研究

【摘要】:目的①觀察動情期非妊娠及植入早期小鼠子宮內(nèi)膜的光鏡結(jié)構(gòu)變化特點;②觀察植入早期小鼠子宮內(nèi)膜Galectin-3和整合素α4的表達(dá)分布狀況;③探討Galectin-3和整合素α4在植入早期小鼠子宮內(nèi)膜表達(dá)的關(guān)系。 方法以成年未經(jīng)產(chǎn)雌性昆明小鼠為研究對象,雌、雄小鼠按3:1合籠過夜,第二天清晨檢查雌鼠陰栓,查到陰栓者記為妊娠第1d。將50只孕鼠隨機(jī)分為5組,每組10只,分別于妊娠1、2、3、4、5d將5組孕鼠頸椎脫臼處死,剖腹取其子宮,另取10只動情期非妊娠小鼠子宮作對照,4%多聚甲醛固定,分別進(jìn)行以下處理:①HE染色觀察小鼠子宮內(nèi)膜的光鏡結(jié)構(gòu)變化;②免疫組化SP法檢測子宮內(nèi)膜Galectin-3和整合素α4的表達(dá)情況;③顯微成像系統(tǒng)拍攝圖像并進(jìn)行計算機(jī)圖像分析系統(tǒng)分析HE染色和免疫組化染色結(jié)果。 結(jié)果①HE染色結(jié)果:動情期非妊娠小鼠子宮內(nèi)膜由腔上皮和固有層組成,固有層中包括子宮腺、基質(zhì)細(xì)胞和血管等,子宮腺為單管狀腺,子宮腔上皮和腺上皮均為單層柱狀上皮。妊娠1~5d小鼠子宮內(nèi)膜呈動態(tài)變化趨勢,主要表現(xiàn)為內(nèi)膜增厚、增生和水腫,妊娠3~5d固有層中子宮腺體數(shù)量漸增多,基質(zhì)細(xì)胞肥大、間隙增大,血管增生。②免疫組化染色結(jié)果顯示:動情期非妊娠及妊娠1~5d小鼠子宮內(nèi)膜腺上皮、腔上皮以及基質(zhì)細(xì)胞中均有Galectin-3的表達(dá)。妊娠1~5d,Galectin-3在妊娠1~2d小鼠子宮內(nèi)膜表達(dá)逐漸增強(qiáng),主要表達(dá)于子宮內(nèi)膜腔上皮和腺上皮,妊娠3d起,表達(dá)明顯增強(qiáng),妊娠4 d、5d呈強(qiáng)表達(dá),妊娠3~5d Galectin-3在基質(zhì)細(xì)胞中有陽性表達(dá),以妊娠4 d、5d表達(dá)最為明顯;動情期非妊娠及妊娠1~5d小鼠子宮內(nèi)膜腺上皮、腔上皮以及基質(zhì)細(xì)胞中均有整合素α4的表達(dá)。妊娠1~5d,整合素α4在妊娠1~3d小鼠子宮內(nèi)膜呈弱陽性表達(dá),主要表達(dá)于子宮內(nèi)膜腔上皮和腺上皮,基質(zhì)細(xì)胞中幾乎不表達(dá)。妊娠4~5d呈強(qiáng)表達(dá),主要表達(dá)于子宮內(nèi)膜腺上皮、腔上皮和基質(zhì)細(xì)胞胞質(zhì)中。③計算機(jī)圖像分析系統(tǒng)計算Galectin-3和整合素α4在妊娠1~5d小鼠子宮內(nèi)膜表達(dá)的平均光密度(MOD)值,經(jīng)SPSS軟件分析,兩者在妊娠1~5d小鼠子宮內(nèi)膜的表達(dá)具有正相關(guān)性。 結(jié)論①妊娠1~5d小鼠子宮內(nèi)膜形態(tài)學(xué)改變呈動態(tài)變化趨勢,以“植入窗”期變化最為明顯;②妊娠1~5d小鼠子宮內(nèi)膜腔上皮、腺上皮和基質(zhì)細(xì)胞的胞質(zhì)中持續(xù)表達(dá)Galectin-3,以妊娠4~5d表達(dá)最強(qiáng),提示Galectin-3可能在小鼠胚胎植入過程中發(fā)揮重要作用;③妊娠1~5d小鼠子宮內(nèi)膜腔上皮、腺上皮和基質(zhì)細(xì)胞的胞質(zhì)中持續(xù)表達(dá)整合素α4,以妊娠4~5d表達(dá)最強(qiáng),提示整合素α4可能參與了小鼠胚胎植入過程;④Galectin-3和整合素α4在妊娠1~5d小鼠子宮內(nèi)膜的表達(dá)具有正相關(guān)性,兩者可能協(xié)同參與了胚胎植入過程。
[Abstract]:Objective 1 to observe the changes of light microscopic structure of endometrium in non-pregnant and early implantation mice during estrous period; 2the expression and distribution of Galectin-3 and integrin 偽 4 in mouse endometrium at the early stage of implantation were observed. 3 to investigate the relationship between Galectin-3 and integrin 偽 4 expression in early implantation of mouse endometrium. Methods female Kunming mice were studied. Female and male mice were treated with 3: 1 cage for the night. The female mice were examined for Yin suppository the next morning. 50 pregnant rats were randomly divided into 5 groups (10 rats in each group). The cervical vertebrae dislocated and the uterus were taken by laparotomy on the 5th day of pregnancy. The uterus of 10 non-pregnant mice in estrous period was fixed with 4% paraformaldehyde. The changes of light microscopic structure of mouse endometrium were observed by the following treatment: 1 he staining. 2Immunohistochemical SP method was used to detect the expression of Galectin-3 and integrin 偽 4 in endometrium. (3) the microscopic imaging system was used to take the images and the computer image analysis system was used to analyze the results of HE staining and immunohistochemical staining. Results 1HE staining showed that the endometrium of non-pregnant mice in estrus was composed of luminal epithelium and lamina propria. The lamina propria included uterine gland, stromal cells and blood vessels, and the uterine gland was single tubular gland. The uterine cavity epithelium and glandular epithelium were monolayer columnar epithelium. On the 3rd and 5th day of pregnancy, the number of uterine glands in lamina propria gradually increased, the stromal cells were hypertrophy and the space was enlarged. The results of immunohistochemical staining showed that the endometrium glandular epithelium of non-pregnant mice during estrous period and that of mice on the 1st and 5th day of gestation. The expression of Galectin-3 was observed in the luminal epithelium and stromal cells, and the expression of Galectin-3 was gradually increased on the 1st and 5th day of gestation in the endometrium of mice on the 1st and 2nd day of gestation. Mainly expressed in endometrial cavity epithelium and glandular epithelium, the expression increased obviously from the 3rd day of gestation, and was strongly expressed on the 4th day of gestation. The positive expression of Galectin-3 was found in stromal cells on the 3rd and 5th day of gestation, especially on the 4th day of gestation. Integrin 偽 4 was expressed in endometrial glandular epithelium, luminal epithelium and stromal cells of non-pregnant and 1-day pregnant mice during estrus. The expression of integrin 偽 4 was weakly positive in the endometrium of mice on the 1st and 3rd day of gestation, mainly in the endometrial cavity epithelium and glandular epithelium, but hardly expressed in stromal cells. Mainly expressed in endometrial glandular epithelium. The mean optical density of Galectin-3 and integrin 偽 4 expression in the endometrium of mice on the 1st day of gestation was calculated by computer image analysis system in the cytoplasm of the lumen epithelium and stromal cells. The results of SPSS software showed that there was a positive correlation between the expression of them in the endometrium of mice on the 1st day of gestation. Conclusion (1) the morphological changes of endometrium in mice on the 1st day of gestation showed a dynamic trend, especially in the period of "implantation window". 2Galectin-3 was continuously expressed in the cytoplasm of endometrial cavity epithelium, glandular epithelium and stromal cells of mice on the 1st day of gestation, and the highest expression was observed on the 5th day of gestation. The results suggest that Galectin-3 may play an important role in mouse embryo implantation. 3 integrin 偽 4 was continuously expressed in the cytoplasm of endometrial cavity epithelium, glandular epithelium and stromal cells of mice on the 1st and 5th day of gestation, and the highest expression was observed on the 5th day of gestation. These results suggest that integrin 偽 4 may be involved in the process of mouse embryo implantation. 4the expression of Galectin-3 and integrin 偽 4 was positively correlated with the expression of integrin 偽 4 in the endometrium of mice on the 1st day of gestation, which may be involved in the process of embryo implantation.
