愈癲湯對(duì)MK-801致精神分裂癥模型大鼠NO信號(hào)通路的影響

【摘要】:目的:基于谷氨酸(Glu)神經(jīng)元功能障礙理論,探討愈癲湯對(duì)地卓西平(MK-801)致精神分裂癥模型大鼠額前腦組織一氧化氮(NO)信號(hào)通路的影響。方法:SD雄性成年大鼠隨機(jī)分為空白組、模型組、對(duì)照組及治療組,每組12只。其中,模型組、對(duì)照組及治療組應(yīng)用0.1mg/m L的MK-801腹腔注射造模,空白組應(yīng)用0.9%氯化鈉溶液行相同方法腹腔注射,連續(xù)14d;于第15天進(jìn)行干預(yù)性給藥,對(duì)照組給予0.036mg/m L利培酮懸濁液灌胃,治療組給予濃度0.92g/m L愈癲湯水煎劑灌胃,空白組、模型組給予相應(yīng)體積蒸餾水灌胃,連續(xù)14d。應(yīng)用分光光度計(jì)法、Western blot法及HPLC-ECD檢測(cè)各組受試大鼠額前腦組織NO活性、一氧化氮合酶(n NOS)蛋白比值及Glu的表達(dá)情況。結(jié)果:治療組受試大鼠額前腦組織NO活性、n NOS比值明顯低于模型組(P0.05),Glu表達(dá)明顯高于模型組(P0.05)。結(jié)論:愈癲湯可以通過抑制NO活性,降低n NOS比值,穩(wěn)定Glu在額前腦組織的表達(dá),從而抑制Glu神經(jīng)毒性,改善Glu神經(jīng)元功能,達(dá)到治療精神分裂癥的目的。
[Abstract]:Objective: based on the theory of glutamate Glu-neuron dysfunction. To investigate the effect of Yuxiangtang on the signal pathway of nitric oxide (no) in frontal forebrain tissue of schizophrenic rats induced by Dizoxipin (MK-801). Methods male adult rats were randomly divided into blank group. Model group, control group and treatment group, 12 rats in each group, model group, control group and treatment group were intraperitoneally injected with 0.1 mg / mL MK-801. The blank group was injected intraperitoneally with 0.9% sodium chloride solution for 14 days. On the 15th day, the control group was given 0.036 mg / mL risperidone suspension, and the treatment group was given 0.92 g / mL Yuxiangshui decoction, blank group. The model group was fed with distilled water of corresponding volume for 14 days. No activity in frontal forebrain of rats in each group was detected by spectrophotometer Western blot and HPLC-ECD. Results: the ratio of nitric oxide synthase (NOS) protein and the expression of Glu were significantly lower in the treatment group than in the model group (P 0.05). The expression of Glu was significantly higher than that of model group P0.050.Conclusion: Yuxiangtang can stabilize the expression of Glu in frontal forebrain by inhibiting the activity of no and decreasing the ratio of n NOS. Thus, the neurotoxicity of Glu was inhibited, the function of Glu neurons was improved, and the treatment of schizophrenia was achieved.
