魚蛋白胨的制備與質(zhì)量分析

【摘要】:目的制備魚蛋白胨,并進(jìn)行質(zhì)量分析。方法將魚糜加工下腳料原料經(jīng)絞碎、石油醚脫脂,得到魚糜漿,用0.1%木瓜蛋白酶酶解,離心收集上清(即魚水解蛋白胨溶液),經(jīng)300 Da納濾膜脫鹽和濃縮,噴霧干燥后,獲得淡黃色粉末,即魚蛋白胨樣品FZU01。對魚蛋白胨FZU01和魚粉蛋白的主要理化指標(biāo)(澄明度、亞硝酸鹽和堿性沉淀、酸堿度、含氮量、氨基氮、干燥失重、熾灼殘渣)進(jìn)行檢測;在固定酵母提取物、Na Cl濃度、初始p H和接種量的基礎(chǔ)上,分別用兩種蛋白胨配制LB培養(yǎng)基,接種大腸埃希菌,檢測A600值;采用分子篩高效液相色譜法(size-exclusion highperformance liquid chromatography,SEC-HPLC)檢測兩種蛋白胨的相對分子質(zhì)量。分別將50、150、300、600、900、1 200和1 500 mg/L組胺以及0.1、0.5、1.0、5.0、10.0、20.0、30.0、40.0和50.0 mg/L重金屬離子(Hg2+、Cd2+、As3+、Cr6+和Pb2+)加至大腸埃希菌中,37℃培養(yǎng)12 h,檢測A600值。結(jié)果兩種蛋白胨理化指標(biāo)檢測結(jié)果均符合《中國生物制品主要原輔材料質(zhì)控標(biāo)準(zhǔn)》規(guī)定,酸堿度、含氮量、氨基氮、干燥失重和熾灼殘渣之間差異無統(tǒng)計學(xué)意義(P0.05),魚蛋白胨FZU01相對分子質(zhì)量主要分布在1 000以內(nèi),占92.5%,魚粉蛋白胨相對分子質(zhì)量主要分布在10 000,小于1 000占56.1%;與魚粉蛋白胨組相比,魚蛋白胨FZU01組生長速率明顯增加(P0.05)。隨著組胺濃度的增加,大腸埃希菌的生長抑制率逐漸增加,與組胺濃度呈正相關(guān),IC50為257.3 mg/L,當(dāng)組胺濃度大于600 mg/L時,完全抑制大腸埃希菌的生長;大腸埃希菌的生長抑制率與重金屬離子濃度呈正相關(guān),Hg~(2+)、Cd~(2+)、As~(3+)、Cr~(6+)和Pb~(2+)對大腸埃希菌的IC50分別為1.35、4.67、7.37、11.04和20.19 mg/L。結(jié)論應(yīng)增加蛋白質(zhì)相對分子質(zhì)量、組胺和重金屬作為蛋白胨產(chǎn)品重要質(zhì)量參數(shù),控制一定的濃度限量范圍,才能保證蛋白胨產(chǎn)品的穩(wěn)定性和安全性。
[Abstract]:Objective to prepare fish peptone and analyze its quality. Methods the raw material of fish surimi was chopped and degreased with petroleum ether, and the surimi was hydrolyzed with 0.1% papain. The supernatant (i.e. fish hydrolyzed peptone solution) collected by centrifugation was desalinated and concentrated by 300Da nanofiltration membrane and then spray-dried to obtain yellowish powder. Fish peptone sample FZU01. The main physicochemical indexes of fish peptone FZU01 and fish meal protein (clarity, nitrite and alkaline precipitation, pH, nitrogen content, amino nitrogen, dry weight loss).
