【摘要】:目的:探討內(nèi)皮克隆形成細(xì)胞條件培養(yǎng)基(ECFCs-CM)對(duì)人真皮成纖維細(xì)胞(HDFs)生物學(xué)作用的影響。方法:從人臍帶血中分離內(nèi)皮克隆形成細(xì)胞(ECFCs)并鑒定;采用抗體芯片檢測(cè)ECFCs-CM中細(xì)胞因子表達(dá)譜;將ECFCs-CM作用于HDFs,EBM-2作為對(duì)照,采用CCK-8法、細(xì)胞劃痕實(shí)驗(yàn)及流式細(xì)胞術(shù)分別檢測(cè)細(xì)胞的增殖、遷移及凋亡情況。結(jié)果:原代培養(yǎng)細(xì)胞符合ECFCs的特征;ECFCs-CM相對(duì)高表達(dá)PDGF-BB、EGF等細(xì)胞因子;實(shí)驗(yàn)組HDFs增殖能力和遷移能力均高于對(duì)照組,且在血清饑餓環(huán)境下實(shí)驗(yàn)組HDFs凋亡率低于對(duì)照組。結(jié)論:ECFCs-CM可促進(jìn)HDFs增殖和遷移,并可抑制其在血清饑餓環(huán)境下的凋亡。
[Abstract]:Objective: to investigate the effect of ECFCs-CM) on human dermal fibroblasts (HDFs). Methods: ECFCswere isolated from human umbilical cord blood and identified. The cytokine expression profile in ECFCs-CM was detected by antibody chip. The proliferation of HDFsN EBM-2 cells was detected by CCK-8 method, cell scratch assay and flow cytometry. Results: the primary cultured cells accorded with the characteristics of ECFCs. The expression of PDGF-BBEGF and other cytokines was relatively high in ECFCs-CM. The proliferative ability and migration ability of HDFs in the experimental group were higher than those in the control group. The apoptosis rate of HDFs in the experimental group was lower than that in the control group under the condition of serum starvation. Conclusion: ECFCs-CM can promote the proliferation and migration of HDFs and inhibit its apoptosis under the condition of serum starvation.
