人臍帶間充質(zhì)干細(xì)胞分離和初步鑒定

【摘要】:目的尋找一種高效獲得穩(wěn)定人臍帶間充質(zhì)干細(xì)胞的方法。方法采集3例足月剖宮產(chǎn)健康胎兒臍帶,剝離血管及外膜得到Wharton’s膠。貼壁培養(yǎng)臍帶Wharton’s膠,收集爬出的細(xì)胞進(jìn)行傳代,觀察細(xì)胞形態(tài);取第3代細(xì)胞采用流式細(xì)胞學(xué)檢測細(xì)胞表面抗原的表達(dá)情況;使用成骨、成脂誘導(dǎo)分化的方法對第3代細(xì)胞進(jìn)行分化潛能的鑒定,成骨誘導(dǎo)細(xì)胞用茜素紅染液染色,成脂誘導(dǎo)細(xì)胞用油紅O染液染色,并與未誘導(dǎo)分化的細(xì)胞進(jìn)行比較。結(jié)果 Wharton’s膠培養(yǎng)10d左右可見梭形細(xì)胞從臍帶組織逸出,貼壁培養(yǎng)細(xì)胞,可見呈放射狀或螺旋狀生長;流式細(xì)胞儀檢測結(jié)果顯示細(xì)胞表面抗原CD29、CD44呈高表達(dá),CD34、CD45呈低表達(dá);成脂誘導(dǎo)培養(yǎng)基中細(xì)胞油紅O染色可見紅色脂滴,成骨誘導(dǎo)培養(yǎng)基中細(xì)胞茜素紅染色可見紅色鈣質(zhì)結(jié)節(jié),未誘導(dǎo)的細(xì)胞未見脂滴及結(jié)節(jié)樣改變。結(jié)論貼壁培養(yǎng)的細(xì)胞符合間充質(zhì)干細(xì)胞生長的形態(tài),有向成骨、成脂分化的能力;貼壁法培養(yǎng)臍帶Wharton膠可得到穩(wěn)定的間充質(zhì)干細(xì)胞。
[Abstract]:Objective to find an efficient stable human umbilical cord mesenchymal stem cells. Methods to collect 3 cases of full-term cesarean section healthy fetal umbilical cord, blood vessels and Wharton membrane stripping 's gel. The adherent culture of umbilical cord Wharton' s glue, climbed out of the collected cells were observed, cell morphology; the third generation the expression of cells by flow cytometry using cell surface antigen; osteogenic differentiation, identification method of adipogenic differentiation potential of the third generation cells, osteogenic cells were stained with alizarin red staining and adipogenic cells induced by oil red O dye staining, and differentiated cells were compared. The Wharton 's Glue 10d training around the spindle cells visible from the umbilical cord tissue escape of cultured adherent cells, visible radially or spiral growth; flow cytometry showed that the cell surface antigen CD29, CD44 showed high expression of CD34, CD45 expression was low; adipogenic cells cultured in medium oil red O staining showed red lipid droplets, osteogenic induction medium cells alizarin red staining showed red calcareous nodules, non induced cells were lipid droplets and nodular changes. Conclusion the adherent cells with mesenchymal stem cell growth morphology have osteogenic, adipogenic differentiation ability; cultured umbilical cord Wharton glue can be obtained by mesenchymal stem cells.
