骨橋蛋白及NF-κB的表達(dá)與大鼠腹主動脈瘤發(fā)生的相關(guān)性

【摘要】:目的構(gòu)建大鼠腹主動脈瘤模型并觀察大鼠腹主動脈瘤中骨橋蛋白(OPN)、核因子-κB(NF-κB)及基質(zhì)金屬蛋白酶-2(MMP-2)的表達(dá)情況,從而探討大鼠腹主動脈瘤發(fā)生的分子機(jī)制。 方法酶灌注法構(gòu)建大鼠腹主動脈瘤模型,首次手術(shù)中測量大鼠腹主動脈直徑,,術(shù)后2周再次手術(shù),測量腹主動脈直徑、取材;HE染色法檢測腹主動脈的結(jié)構(gòu)改變及炎性細(xì)胞浸潤情況;特殊染色法檢測腹主動脈中層彈力蛋白變化情況;免疫組化技術(shù)、WesternBlot法檢測各組腹主動脈組織中OPN、NF-κB及基質(zhì)金屬蛋白酶-2(MMP-2)的表達(dá)量。 結(jié)果成功構(gòu)建大鼠腹主動脈瘤模型;腹主動脈瘤組術(shù)后直徑擴(kuò)張率(220.3%)明顯高于對照組(112.3%);HE染色結(jié)果顯示腹主動脈瘤組動脈壁中各層結(jié)構(gòu)紊亂,炎性細(xì)胞浸潤程度顯著增高;維多利亞特殊染色顯示腹主動脈瘤組動脈壁有不同程度的彈力纖維消失,萎縮膠原纖維量大量增加;免疫組化結(jié)果顯示OPN在腹主動脈瘤組、對照組及正常組動脈壁中的表達(dá)分別為7.5×1012±2.92×1011、3.07×10~5±2.58×10~5、0(F=6.59,P=0.004);NF-κB在腹主動脈瘤組、對照組及正常組動脈壁中表達(dá)量分別為2.36×10~(10)±2.80×10~(10)、0、0(F=6.96,P=0.004);MMP-2在腹主動脈瘤組、對照組及正常組動脈壁中的表達(dá)量分別是4.52×10~(10)±1.73×10~(10)、0、0(F=6.83,P=0.003);WesternBlot結(jié)果顯示OPN在腹主動脈瘤組、對照組及正常組動脈壁中的表達(dá)分別為2.77×10~5±2.82×10~5、4.52×10~3±1.36×10~3、2.11×10~3±1.99×10~3(F=9.49,P=0.001);NF-κB在腹主動脈瘤組、對照組及正常組動脈壁中表達(dá)量分別為6.014×10~4±6.27×10~4、1.34×10~3±1.89×10~3、4.35×10~2±7.01×10~2(F=8.91,P=0.001);MMP-2在腹主動脈瘤組、對照組及正常組動脈壁中的表達(dá)量分別是9.25×10~4±6.87×10~4、8.00×10~2±7.89×10~2、2.70×10~2±3.89×10~2(F=17.91,P㩳0.001),OPN、NF-κB及基質(zhì)金屬蛋白酶-2(MMP-2)呈正相關(guān)。 結(jié)論成功構(gòu)建大鼠腹主動脈瘤模型,骨橋蛋白、NF-κB及MMP-2與大鼠腹主動脈瘤形成存在相關(guān)性,骨橋蛋白可能通過NF-κB途徑上調(diào)MMP的表達(dá),過度表達(dá)的MMP加速了動脈壁中細(xì)胞外基質(zhì)的降解而在大鼠腹主動脈瘤的發(fā)生發(fā)展中起到重要作用。
[Abstract]:Objective to establish a rat model of abdominal aortic aneurysm and to observe the expression of osteopontin (OPN), nuclear factor- 魏 B (NF- 魏 B) and matrix metalloproteinase-2 (MMP-2) in abdominal aortic aneurysm. To explore the molecular mechanism of abdominal aortic aneurysm in rats. Methods A rat model of abdominal aortic aneurysm was established by enzyme perfusion. The diameter of abdominal aorta was measured in the first operation. The changes of abdominal aorta structure and inflammatory cell infiltration were detected by HE staining. The changes of elastin in abdominal aorta were detected by special staining method. The expression of OPN- 魏 B and matrix metalloproteinase-2 (MMP-2) in abdominal aorta was detected by Western blot. Results A rat model of abdominal aortic aneurysm was successfully established. The diameter expansion rate of abdominal aortic aneurysm group (220.3%) was significantly higher than that of control group (112.3%). The results of HE staining showed that in the abdominal aortic aneurysm group, the structure of each layer was disordered, and the degree of inflammatory cell infiltration was significantly increased. Victorian special staining showed that elastic fibers disappeared and the amount of atrophy collagen fibers increased significantly in abdominal aortic aneurysm group. There was a positive correlation between NF- 魏 B and matrix metalloproteinase-2 (MMP-2). Conclusion A rat model of abdominal aortic aneurysm was successfully established. Osteopontin NF- 魏 B and MMP-2 were correlated with the formation of abdominal aortic aneurysm in rats. Osteopontin may up-regulate the expression of MMP through NF- 魏 B pathway. Overexpression of MMP accelerates the degradation of extracellular matrix in arterial wall and plays an important role in the development of abdominal aortic aneurysm in rats.
