【摘要】：背景:針灸對(duì)腦性癱瘓幼鼠有一定的治療作用。目的:探討火針對(duì)腦性癱瘓幼鼠腦組織中氨基酸和神經(jīng)遞質(zhì)表達(dá)的影響。方法:取1周齡Wistar幼鼠50只,45只結(jié)扎左側(cè)頸總動(dòng)脈建立幼鼠腦性癱瘓模型,5只不處理作為對(duì)照組。45只模型幼鼠隨機(jī)分為模型組、針刺組及陽(yáng)性組,每組15只。于造模第3天開(kāi)始,陽(yáng)性組灌胃腦酶水解片溶液,火針組使用火針處理內(nèi)關(guān)穴、涌泉穴、曲池穴、百會(huì)穴,治療持續(xù)21 d。于末次治療1 h后,進(jìn)行懸掛實(shí)驗(yàn)及Y型迷宮實(shí)驗(yàn),并處死大鼠分離其大腦皮質(zhì)及海馬區(qū),組織勻漿后檢測(cè)乙酰膽堿、去甲腎上腺素、5-羥色胺、谷氨酸和γ-氨基丁酸的表達(dá)變化,采用病理切片觀察海馬區(qū)組織結(jié)構(gòu)變化。結(jié)果與結(jié)論:(1)一般情況:21 d內(nèi)治療組大鼠活動(dòng)明顯多于模型組。治療21 d,與對(duì)照組相比,模型組體質(zhì)量顯著下降(P0.05);與模型組相比,陽(yáng)性組及火針組體質(zhì)量均明顯上升(P0.05),且后2組差異無(wú)顯著性意義;(2)懸掛實(shí)驗(yàn)及Y型迷宮實(shí)驗(yàn):與對(duì)照組相比,模型組懸掛時(shí)間顯著下降,犯錯(cuò)次數(shù)顯著性上升(P0.05);與模型組相比,陽(yáng)性組及火針組懸掛時(shí)間顯著性延長(zhǎng),陽(yáng)性組和火針組的犯錯(cuò)次數(shù)顯著下降(P0.05);(3)神經(jīng)遞質(zhì)水平:與對(duì)照組相比,模型組去甲腎上腺素、5-羥色胺和γ-氨基丁酸水平下降,谷氨酸水平增加(P0.05);與模型組相比,陽(yáng)性組和火針組的去甲腎上腺素、5-羥色胺和γ-氨基丁酸水平增加,谷氨酸水平減少(P0.05);(4)蘇木精-伊紅染色:模型組可見(jiàn)大量的神經(jīng)元結(jié)構(gòu)破壞變形,體積減小,細(xì)胞核固縮,且與細(xì)胞膜邊界不清晰;陽(yáng)性組和火針組都可以在一定程度上治療腦缺血引起的海馬區(qū)腦細(xì)胞結(jié)構(gòu)異常。(5)結(jié)果證明,火針?lè)ǹ梢杂行е委熡资竽X性癱瘓。
[Abstract]:Background: acupuncture has a certain therapeutic effect on cerebral palsy young rats. Objective: to investigate the effect of fire on the expression of amino acids and neurotransmitters in brain tissue of cerebral palsy young rats. Fifty one-week-old Wistar rats were selected. 45 young rats with cerebral palsy were randomly divided into three groups: model group, acupuncture group and positive group. 15 rats in each group were treated with Neiguan points, Yongquan points, Quchi points and Baihui points in the positive group after the 3rd day of model making. After the last treatment for 1 hour, suspension test and Y-maze test were carried out. The rats were killed to isolate their cerebral cortex and hippocampus, and acetylcholine and norepinephrine were detected after tissue homogenate. Changes in the expression of 5-hydroxytryptamine, glutamate and 緯 -aminobutyric acid. Results and conclusion the activity of rats in the treatment group was significantly higher than that in the model group within 21 days after treatment, and was compared with that in the control group. The body mass of the model group decreased significantly (P 0.05). Compared with the model group, the body weight of the positive group and the fire needle group were significantly higher than that of the model group, and there was no significant difference between the latter two groups. (2) suspension test and Y-maze test: compared with the control group, the suspension time of the model group decreased significantly, and the number of mistakes increased significantly (P 0.05). Compared with the model group, the suspension time of the positive group and the fire needle group was significantly prolonged, and the number of mistakes in the positive group and the fire needle group decreased significantly (P 0.05). (3) level of neurotransmitters: compared with the control group, the levels of norepinephrine 5-hydroxytryptamine and 緯 -aminobutyric acid in the model group decreased, while the level of glutamate increased (P 0.05). Compared with the model group, the levels of norepinephrine 5-hydroxytryptamine and 緯 -aminobutyric acid in the positive group and the fire needle group were increased, and the level of glutamate was decreased (P 0.05). (4) hematoxylin-eosin staining: in the model group, a large number of neuronal structures were destroyed and deformed, the volume decreased, the nucleus was pyknosis, and the boundary with the cell membrane was not clear. Both the positive group and the fire acupuncture group could treat the abnormal structure of hippocampal cells caused by cerebral ischemia to a certain extent. The results showed that the fire acupuncture method could effectively treat cerebral palsy in young rats.
