【摘要】：目的:采用腹腔注射鏈脲佐菌素(STZ)法復(fù)制糖尿病大鼠模型,觀察糖尿病模型大鼠不同時(shí)期視網(wǎng)膜組織中基質(zhì)金屬蛋白酶2(MMP-2)表達(dá)水平,闡明MMP-2在糖尿病大鼠糖尿病視網(wǎng)膜病變(DR)中的作用。方法:雌性SD大鼠分為正常對照組(n=24)、4周模型組(n=30)、6周模型組(n=30)和8周模型組(n=30),模型組大鼠連續(xù)5d腹腔注射小劑量STZ復(fù)制糖尿病大鼠模型,正常對照組大鼠注射等量的檸檬酸鈉溶液。4、6和8周時(shí)檢測各組大鼠體質(zhì)量和血糖水平。末次檢測后麻醉獲取大鼠右眼視網(wǎng)膜組織,采用RT-PCR和Western blotting法檢測MMP-2mRNA及蛋白表達(dá)水平,免疫組織化學(xué)染色觀察大鼠視網(wǎng)膜組織中MMP-2表達(dá)情況。結(jié)果:造模前,各組大鼠體質(zhì)量和禁食12h的空腹血糖值比較差異均無統(tǒng)計(jì)學(xué)意義(P0.05)。4周模型組大鼠死亡3只,6周時(shí)死亡5只,8周時(shí)死亡7只,存活動(dòng)物繼續(xù)進(jìn)行實(shí)驗(yàn)。各模型組大鼠體質(zhì)量均明顯低于同時(shí)期正常對照組(P0.05或P0.01),而空腹血糖值明顯升高(P0.05或P0.01)。各模型組大鼠視網(wǎng)膜組織中MMP-2mRNA和蛋白表達(dá)水平均明顯高于正常對照組(P0.05)。正常對照組大鼠視網(wǎng)膜結(jié)構(gòu)層次清晰分明、細(xì)胞排列整齊,可見單層排列的神經(jīng)節(jié)細(xì)胞,核圓形或橢圓形狀,核較大;與正常對照組比較,模型組大鼠視網(wǎng)膜組織結(jié)構(gòu)松散,內(nèi)核層及視桿細(xì)胞層細(xì)胞外界膜模糊,數(shù)量減少。MMP-2陽性細(xì)胞可見棕黃色顆粒沉淀,在神經(jīng)節(jié)細(xì)胞層胞漿和血管內(nèi)皮細(xì)胞尤為集中。4、6及8周模型組MMP-2陽性表達(dá)水平均高于正常對照組(P0.05)。結(jié)論:MMP-2在糖尿病模型大鼠視網(wǎng)膜上表達(dá),并隨著糖尿病大鼠模型維持時(shí)間的延長呈遞增趨勢,說明MMP-2與DR有關(guān)。
[Abstract]:Objective: by intraperitoneal injection of streptozotocin (STZ) diabetic rat model was established to observe matrix metalloproteinases in retinal tissue of diabetic rats in different periods in 2 (MMP-2) expression level, clarify the MMP-2 in diabetic rats with diabetic retinopathy (DR) in rats. Methods: female SD rats were divided into the normal control group (n=24), 4 week model group (n=30), model group (n=30) for 6 weeks and 8 weeks model group (n=30), model group rats 5D continuous intraperitoneal injection of small dose STZ model of diabetic rats, normal control of sodium citrate solution in.4,6 rats injected and 8 weeks when the rats were measured body weight and blood glucose level. At the time of detection after anesthesia for the right eye retina of rats, using RT-PCR and Western blotting method to detect the expression of MMP-2mRNA and protein expression of MMP-2 in retinal tissues of rats with immunohistochemical staining: Before modeling, the body weight and fasting 12h rats fasting glucose showed no significant difference (P0.05).4 weeks rats 3 rats died in 6 weeks, 5 rats died in 8 weeks, 7 died and the survival of the animal to continue the experiment. The body weight of rats in model group were significantly at the same time lower than the normal control group (P0.05 or P0.01), and fasting blood glucose increased significantly (P0.05 or P0.01). The level was significantly higher than that of normal control group and the expression of MMP-2mRNA protein in retinal tissue of rats in the model group (P0.05). Normal control group rat retinal structure level clearly, cells arranged in neat, visible monolayer ganglion cells, nucleus round or oval shape, a large nucleus; compared with normal control group, the loose structure of retinal tissues of rats in the model group, the kernel layer and rod cell layer cell membrane outside fuzzy, reduce the number of.MMP-2 positive cells showed Brown particles sink The lake, in the ganglion cell layer of cytoplasm and endothelial cells is particularly concentrated.4,6 and 8 weeks model group MMP-2 expression levels were higher than normal control group (P0.05). Conclusion: the expression of MMP-2 in retina of diabetic rats, and with the model of diabetic rats maintain time increasing, MMP-2 and DR.
