一種酶抗逆助劑的制備及其在廢水凈化處理中應(yīng)用

【摘要】:近海岸海水、船舶廢水(壓載水、洗艙水、脫硫廢液)等污廢水中均含有較高濃度的氨氮和石油烷烴。對這類廢水的生物凈化處理,要求具有高鹽適應(yīng)性、能同時(shí)降解轉(zhuǎn)化氨氮和石油烷烴的特性及高效性。本文擬利用抗逆助劑協(xié)助下的復(fù)合酶制劑污廢水凈化處理技術(shù),進(jìn)行模擬船舶廢水脫氮和石油烷烴降解研究。本文從大連某鹽場的鹽池底泥中分離篩選出一株能夠合成Ectoine的菌株H02,菌株H02經(jīng)形態(tài)鑒定、16S rDNA鑒定為Halomonas屬菌株,命名為Halomonas sp.H02。利用液相色譜與紅外光譜特征峰分析的方法鑒定Halomonas sp.H02合成物質(zhì),確定其為Ectoine。經(jīng)分泌性鑒定Halomonassp.H02為Ectoine非分泌型菌株?疾炝薍alomonas sp.H02的的生長及Ectoine發(fā)酵特性,其最適發(fā)酵培養(yǎng)基種類為MG改良培養(yǎng)基,最適碳源種類為谷氨酸鈉,最適鹽濃度為90 g/L;利用單因素實(shí)驗(yàn)考察了影響Ectoine合成的培養(yǎng)基各因素水平,并通過響應(yīng)面法尋求最優(yōu)的Ectoine發(fā)酵培養(yǎng)基,采用低滲透壓沖擊法制備Ectoine,所得制備率均值為94.0%?疾炝死备^氧化物酶(HRP)和漆酶兩種污水處理常用酶的最適測定條件及Ectoine對極端環(huán)境下兩種酶的的抗逆保護(hù)作用。結(jié)果表明:HRP酶的最適溫度為30 ℃,最適pH為6,NaCl濃度對HRP酶活性具有明顯抑制作用;漆酶對鹽敏感,最適溫度為40 ℃,最適pH為4。Ectoine對于高鹽、極端溫度、極端pH下酶活性的保護(hù)具有顯著作用。將HRP、漆酶及其它具有降解石油、脫氮功能性的細(xì)胞粗酶干粉混合制備酶制劑,同時(shí)添加Ectoine作為抗逆助劑。結(jié)果表明:混合酶抗逆助劑對于模擬船舶含油污水(含油脫硫廢液)具有顯著處理效果,石油降解率最高達(dá)到52.5%,此時(shí)脫氮率為44.2%。
[Abstract]:High concentrations of ammonia nitrogen and petroleum alkanes are found in marine wastewater (ballast water, tank washing water, desulphurization waste) and so on. The biological purification and treatment of this kind of wastewater requires high salt adaptability, the characteristics and high efficiency of simultaneous degradation of ammonia nitrogen and petroleum alkanes. In this paper, the removal of nitrogen from simulated ship wastewater and the degradation of petroleum alkanes are studied by using the technology of purification and treatment of wastewater from wastewater with complex enzyme preparation assisted by anti-stress auxiliaries. A strain H02 capable of synthesizing Ectoine was isolated from the sediment of a salt pond in a salt field in Dalian. The strain H02 was identified as Halomonas genus by morphological identification of 16s rDNA and named Halomonas sp. H02. The synthetic compound of Halomonas sp.H02 was identified as Ectoine by liquid chromatography and infrared spectrum characteristic peak analysis. Halomonassp.H02 was identified as a non-secretory Ectoine strain by secretion. The growth of Halomonas sp.H02 and the characteristics of Ectoine fermentation were investigated. The optimum fermentation medium was MG improved medium and the most suitable carbon source was sodium glutamate. The optimum concentration of salt was 90 g / L, the factors affecting Ectoine synthesis were investigated by single factor experiment, and the optimal Ectoine fermentation medium was obtained by response surface method, and Ectoine was prepared by low osmotic pressure impact method.
