鄰苯二甲酸二甲酯暴露對凡納濱對蝦殼膜幾丁質酶的影響

【摘要】:以凡納濱對蝦為試驗對蝦,通過不同濃度0、2、10、20μg/L鄰苯二甲酸二甲酯(DMP)暴露不同時間(120、240 h)后,采用酶促反應動力學法及實時PCR檢測分析對蝦殼膜幾丁質酶比活力及幾丁質酶mRNA表達量差異。結果顯示,DMP暴露對對蝦殼膜幾丁質酶比活力具有抑制作用,但不具有顯著性。不同濃度DMP暴露不同時間后,對對蝦殼膜幾丁質酶系列基因的mRNA表達量均受到不同程度的影響,2μg/L DMP暴露120 h時,對蝦殼膜Lv CHT3mRNA表達量顯著上升;2μg/L DMP暴露240 h時,Lv CHT2 mRNA表達量顯著下降;20μg/L DMP暴露240 h時,Lv CHT6 mRNA表達量也顯著下降,說明DMP對對蝦生長發(fā)育具有毒性。
[Abstract]:Using Penaeus vannamei as an experimental shrimp, after being exposed to DMP20 渭 g / L of dimethyl phthalate for 120240 h at different concentrations, The specific activity of chitinase and the difference of chitinase mRNA expression in shrimp shell membrane were analyzed by enzymatic kinetic method and real-time PCR detection. The results showed that the specific activity of chitinase in shrimp shell membrane was inhibited by DMP exposure, but it was not significant. After exposure to different concentrations of DMP for different time, the mRNA expression of chitinase genes in shrimp shell was affected by different degrees of exposure to 2 渭 g / L DMP for 120h. The expression of Lv CHT3mRNA in shrimp shell membrane was significantly increased with 2 渭 g / L DMP exposure for 240h. The expression of Lv CHT2 mRNA in shrimp shell membrane decreased significantly when exposed to 20 渭 g / L DMP for 240h, indicating that DMP is toxic to the growth and development of Penaeus penaeus.
