背景:胃癌作為一種上消化道惡性腫瘤,具有較高的發(fā)病率和死亡率。由于胃癌發(fā)病初期無明顯癥狀,很多患者確診時(shí)疾病已發(fā)展到進(jìn)展期,給后續(xù)治療加大了難度。目前,針對(duì)胃癌的主要治療手段仍是手術(shù)和化療,然而大部分進(jìn)展期胃癌患者經(jīng)過治療后仍會(huì)發(fā)生復(fù)發(fā)和轉(zhuǎn)移,導(dǎo)致進(jìn)展期胃癌患者的預(yù)后較差,全球范圍內(nèi)整體5年生存率僅為29%。雖然在傳統(tǒng)治療策略的基礎(chǔ)上,許多新的治療方法已被發(fā)現(xiàn),并應(yīng)用于胃癌治療,然而這些治療策略在進(jìn)展期胃癌患者中的治療效果并不理想。改善胃癌患者的預(yù)后生存,明確胃癌轉(zhuǎn)移機(jī)制是當(dāng)今胃癌研究的重點(diǎn)。隨著對(duì)胃癌研究的不斷深入,人們開始從基因?qū)用嫣骄课赴┑陌l(fā)生及發(fā)展并發(fā)現(xiàn)了許多胃癌相關(guān)的突變基因及信號(hào)通路,這些發(fā)現(xiàn)促進(jìn)了分子靶向治療的興起。靶向EGFR的西妥昔單抗及靶向VEGFR-2的阿帕替尼在改善進(jìn)展期胃癌患者的預(yù)后生存方面的成功應(yīng)用,彰顯了分子靶向治療的重要性。分子靶向治療的成功有賴于靶向胃癌的高敏感度藥物的研究及新的藥物靶點(diǎn)的篩選。針對(duì)胃癌相關(guān)差異基因的分析可以篩選出胃癌關(guān)鍵基因,進(jìn)一步對(duì)關(guān)鍵基因表達(dá)及生物學(xué)功能的研究將為靶點(diǎn)的搜尋提供強(qiáng)大的助力。過表達(dá)的FBN1在細(xì)胞增殖、細(xì)胞凋亡和腫瘤...

吉林大學(xué)博士學(xué)位論文2.3.3 差異表達(dá) miRNA 靶標(biāo)基因預(yù)測(cè)利用 miRWalk 3.0 數(shù)據(jù)庫對(duì)差異表達(dá) miRNA 的靶標(biāo)基因進(jìn)行預(yù)測(cè)。26 個(gè)差異表達(dá) miRNA 一共靶向 630 個(gè)靶標(biāo)基因，根據(jù) miRWalk 3.0 預(yù)測(cè)工具繪制出 miRNA與靶標(biāo)基因的互作網(wǎng)絡(luò)關(guān)系圖（圖 2.4）。
