超積累型東南景天Sa12F279基因的抗逆表達(dá)響應(yīng)及功能關(guān)聯(lián)分析

[目的]對(duì)超積累型東南景天ABC轉(zhuǎn)運(yùn)蛋白家族成員Sa12F279開展生物信息學(xué)分析及表達(dá)分析,為探究ABC類轉(zhuǎn)運(yùn)蛋白在鎘、干旱、鹽堿等非生物脅迫中的功能提供參考。[方法]通過對(duì)超積累型東南景天轉(zhuǎn)錄組數(shù)據(jù)庫進(jìn)行比對(duì)分析,獲得1個(gè)ABC家族成員的基因。通過生物信息學(xué)方法分析Sa12F279基因的進(jìn)化關(guān)系、蛋白結(jié)構(gòu)域構(gòu)成、核心結(jié)構(gòu)域同源比對(duì)及在組學(xué)數(shù)據(jù)中相關(guān)的互作蛋白分類;借助實(shí)時(shí)定量PCR技術(shù)分析該基因在鹽堿、干旱及ABA激素脅迫下根中的表達(dá)變化。[結(jié)果]通過比對(duì)分析獲得ABC家族成員Sa12F279基因,該基因的開放閱讀框長(zhǎng)度為4 497 bp,編碼蛋白長(zhǎng)度為1 498個(gè)氨基酸,相對(duì)分子量為167.1 KD,等電點(diǎn)為6.92。進(jìn)化分析顯示:Sa12F279基因與C亞類成員聚為一簇,且在結(jié)構(gòu)域構(gòu)成上符合ABC家族的保守排布。共表達(dá)網(wǎng)絡(luò)分析顯示:Sa12F279與567個(gè)基因存在功能上的關(guān)聯(lián),對(duì)這些基因進(jìn)行功能注釋發(fā)現(xiàn),其中39.8%的基因執(zhí)行代謝相關(guān)功能,29.3%的基因與細(xì)胞內(nèi)進(jìn)程相關(guān),10.2%的基因涉及生物調(diào)控,7.1%的基因參與轉(zhuǎn)運(yùn)活性。對(duì)鎘脅迫前后轉(zhuǎn)錄組數(shù)據(jù)分析顯示,Sa12F2...
