小麥凝集素激酶TaLecRK-Ⅳ.1基因的作用和由紋枯病誘導(dǎo)的小麥基因發(fā)展的分子標(biāo)記

  小麥紋枯病主要由土傳真菌Rhizoctonia cerealis侵染小麥莖基部引起。它已經(jīng)成為世界范圍內(nèi)小麥生產(chǎn)的限制性因素,在中國(guó)更為嚴(yán)重。小麥紋枯病的防治已經(jīng)成為小麥生產(chǎn)可持續(xù)發(fā)展的主要問(wèn)題。培育和種植抗性小麥品種是防治小麥紋枯病最環(huán)保、高效的方法。小麥紋枯病抗性遺傳機(jī)制研究有助于解析小麥抗性相關(guān)基因的功能和作用。本研究中,我們從抗紋枯病的小麥材料CI12633中鑒定了一個(gè)L型凝集素受體激酶基因,命名為T(mén)aLecRK-Ⅳ.1。與感紋枯病小麥溫麥6號(hào)相比,在CI12633中該基因的表達(dá)受紋枯病侵染誘導(dǎo)。序列分析表明,該基因編碼含有676個(gè)氨基酸殘基的L型凝集素受體激酶。利用病毒誘導(dǎo)基因沉默技術(shù)(virus-induced gene silencing,VIGS)技術(shù)沉默CI12633的TaLecRK-Ⅳ.1后,小麥植株表現(xiàn)矮化的表型,表明該基因可能是小麥矮化的負(fù)調(diào)控因子。進(jìn)一步在含有矮化基因Rht-D1b(formerly Rht2)的小麥中分析了該基因的轉(zhuǎn)錄,結(jié)果表面TaLecRK-Ⅳ.1在矮化小麥中的表達(dá)量很低。另外,參與植物生長(zhǎng)和發(fā)育的激素---赤霉素和生長(zhǎng)素誘導(dǎo)TaLecRK...
