水稻低溫應(yīng)答基因OsLCL3的分離與功能鑒定

【摘要】:為探明水稻響應(yīng)低溫的分子機(jī)制,改良水稻對(duì)低溫的抗性,從水稻低溫誘導(dǎo)表達(dá)譜中挑選了1個(gè)受低溫誘導(dǎo)的基因OsLCL3,通過(guò)實(shí)時(shí)定量PCR和啟動(dòng)子驅(qū)動(dòng)GUS報(bào)告基因轉(zhuǎn)化水稻的方法驗(yàn)證其功能。對(duì)OsLCL3進(jìn)行同源蛋白搜索,發(fā)現(xiàn)它屬于DUF761超家族,并對(duì)這個(gè)超家族中的部分蛋白序列進(jìn)行了系統(tǒng)樹分析;通過(guò)水稻原生質(zhì)體瞬時(shí)表達(dá)系統(tǒng),初步確定OsLCL3蛋白定位于細(xì)胞核和細(xì)胞質(zhì)中。在水稻中超量表達(dá)OsLCL3基因,并對(duì)轉(zhuǎn)基因植株進(jìn)行低溫脅迫處理,發(fā)現(xiàn)超量表達(dá)OsLCL3的植株在低溫脅迫下的電導(dǎo)率顯著低于野生型對(duì)照植株,脅迫后超量表達(dá)植株的存活率顯著高于野生型對(duì)照植株。結(jié)果表明,OsLCL3可能在水稻對(duì)低溫的應(yīng)答中起著重要作用。
[Abstract]:In order to find out the molecular mechanism of rice response to low temperature and improve the resistance of rice to low temperature, a low temperature induced gene OsLCL3, was selected from the low temperature induction expression profile of rice to verify its function by real time quantitative PCR and promoter driven GUS reporter gene transformation into rice. The homologous protein search of OsLCL3 was carried out, and it was found that it belonged to DUF761 superfamily, and some protein sequences of DUF761 superfamily were analyzed by systematic tree analysis, and the localization of OsLCL3 protein in nucleus and cytoplasm was preliminarily determined by transient expression system of rice protoplasts. The OsLCL3 gene was overexpressed in rice and the transgenic plants were treated with low temperature stress. It was found that the electrical conductivity of the plants overexpressing OsLCL3 under low temperature stress was significantly lower than that of the wild type control plants, and the survival rate of the plants overexpressed after stress was significantly higher than that of the wild type control plants. The results showed that OsLCL3 may play an important role in the response of rice to low temperature.
