連翹2個(gè)DIR基因的分子特征與表達(dá)分析
[Abstract]:Objective: to clone Forsythia suspensa DIR gene and analyze its molecular characteristics and expression characteristics. Methods: two full-length sequences of Forsythia suspensa DIR gene were cloned and their expression in different tissues of Forsythia suspensa and induced by methyl jasmonate was analyzed by real-time fluorescence quantitative PCR. Results: two DIR genes (FsDIR1,MF168408;FsDIR2,MF168409) were cloned and the amino acids encoded by 558 bp and 564 bp, were 185 and 187, respectively. Phylogenetic tree analysis showed that FsDIR1 belonged to DIR-a subfamily and FsDIR2 belonged to DIR-b/d subfamily. The results of real-time fluorescence quantitative PCR in different tissues of Forsythia suspensa showed that the expression of FsDIR1 and FsDIR2 was the lowest in petals and the highest in pistil. After induction with methyl jasmonate, the contents of phillyrin and phillyrin A in leaves increased, but the expression of FsDIR1 and FsDIR2 decreased significantly. Conclusion: two DIR genes of Forsythia suspensa were cloned in this study, which laid a foundation for studying the function of DIR gene in the biosynthesis of Forsythia suspensa lignans and its involvement in plant resistance.
