西昌市雞蛔蟲(chóng)線粒體cyt b和nad1基因的遺傳變異分析

【摘要】:為研究西昌市雞蛔蟲(chóng)的種內(nèi)遺傳變異和系統(tǒng)發(fā)生關(guān)系,用線粒體細(xì)胞色素b(cyt b)和煙酰胺腺嘌呤二核苷酸脫氫酶亞單位Ⅰ(nad1)基因全序列分析了17個(gè)雞蛔蟲(chóng)西昌分離株的遺傳變異,并用cyt b和nad1序列構(gòu)建雞蛔蟲(chóng)與其他蛔蟲(chóng)的種系發(fā)育關(guān)系。測(cè)序結(jié)果顯示:17株雞蛔蟲(chóng)cyt b和nad1全序列長(zhǎng)度分別為1 104 bp和876 bp,堿基變異率分別為02.4%和02.3%;分別檢測(cè)到10個(gè)和7個(gè)單倍型,總的單倍型多樣性分別為0.897±0.056和0.824±0.064,核苷酸多樣性分別為0.004 70±0.001 61和0.004 21±0.001 65,平均遺傳距離分別為0.005和0.004。構(gòu)建的種系發(fā)育樹(shù)均顯示17個(gè)雞蛔蟲(chóng)西昌分離株與其他國(guó)家/地區(qū)的雞蛔蟲(chóng)分離株聚類形成一個(gè)分支,與鴿蛔蟲(chóng)的親緣關(guān)系最近,與其他蛔蟲(chóng)所屬的分支相隔較遠(yuǎn)。研究結(jié)果表明雞蛔蟲(chóng)西昌分離株的遺傳變異程度低,且cyt b基因比nad1基因更適合作為研究雞蛔蟲(chóng)種內(nèi)遺傳變異的分子標(biāo)記。
[Abstract]:In order to study the intraspecific genetic variation and phylogenetic relationship of Ascaris lumbricoides in Xichang City, the genetic variation of 17 Ascaris lumbricoides isolates was analyzed by using the complete sequence of b (cyt b) and nad1 genes, and the relationship between Ascaris lumbricoides and other Ascaris lumbricoides was constructed by cyt b and nad1 sequences. The sequencing results showed that the full sequence length of cyt b and nad1 of 17 strains of Ascaris lumbricoides were 1.104 bp and 0.2.3%, 10 and 7 haplotypes were detected, the total haplotype diversity was 0.897 鹵0.056 and 0.824 鹵0.064, the nucleotides diversity was 0.004 70 鹵0.001 61 and 0.004 21 鹵0.001 65, and the average genetic distance was 0.005 and 0.004, respectively. The constructed species development trees showed that 17 isolates of Ascaris lumbricoides Xichang formed a branch with the isolates of Ascaris lumbricoides in other countries and regions, which were closely related to Ascaris pigeon and far from the branches of other Ascaris lumbricoides. The results showed that the genetic variation of Ascaris lumbricoides Xichang isolate was low, and cyt b gene was more suitable than nad1 gene as a molecular marker to study the intraspecific genetic variation of Ascaris lumbricoides.
