人工基因元件的生理學(xué)研究進展

【摘要】:按照工程學(xué)原理人工設(shè)計的基因元件應(yīng)該是模塊化的,同時具備可預(yù)測地組裝和再利用的屬性。然而,真正的細胞生理條件下各種層次的生理干涉效應(yīng)會嚴重地阻礙人工基因元件的功能性組裝,即大多數(shù)組裝后的人工系統(tǒng)完全或部分喪失預(yù)設(shè)功能。我們提出合成生理學(xué)的概念,將其定義為研究和控制人工生命系統(tǒng)與底盤細胞生理系統(tǒng)相互作用的合成生物學(xué)分支領(lǐng)域。在此框架下,本文歸納了細胞生理系統(tǒng)與人工基因元件的相互干涉方式,并對表征和消除這種相互作用的技術(shù)方法和設(shè)計策略進行綜述。
[Abstract]:Genetic elements designed artificially according to engineering principles should be modular and have predictable assembly and reuse properties. However, under the real cell physiological conditions, the physiological interference effects at various levels will seriously hinder the functional assembly of artificial gene elements, that is, most of the assembled artificial systems completely or partially lose their preset function. We propose the concept of synthetic physiology, which is defined as a branch of synthetic biology to study and control the interaction between artificial life system and chassis cell physiological system. In this framework, the interference between cell physiological system and artificial gene elements is summarized, and the technical methods and design strategies to characterize and eliminate this interaction are reviewed.
