豬流行性腹瀉病毒山西分離株S基因遺傳變異分析

【摘要】:為分析山西地區(qū)豬流行性腹瀉病毒(PEDV)的遺傳變異情況,試驗(yàn)利用RT-PCR方法對(duì)2014-2015年山西省疑似豬流行性腹瀉的陽(yáng)性病料進(jìn)行克隆和測(cè)序,獲得4個(gè)S基因片段,并對(duì)其基因序列和推導(dǎo)的氨基酸序列與國(guó)內(nèi)外毒株進(jìn)行比對(duì)分析。序列分析結(jié)果顯示,4株P(guān)EDV山西分離株的S基因與CV777vaccine相比,在170~171bp之間插入12個(gè)核苷酸,在401~402、454~455bp之間均插入3個(gè)核苷酸,在461~468bp之間缺失6個(gè)核苷酸。4株P(guān)EDV山西分離株S基因之間核苷酸和氨基酸同源性分別為99.2%~99.8%和98.6%~99.7%,與2011-2015年中國(guó)流行毒株、CV777vaccine、attenuated DR13、CV777的核苷酸同源性分別95.0%~98.5%、93.2%~93.6%、93.2%~93.7%、93.7%~94.4%,氨基酸同源性分別為96.2%~98.9%、91.9%~92.6%、92.1%~92.9%、92.9%~94.0%。遺傳進(jìn)化樹分析結(jié)果表明,PEDVS基因分為3個(gè)群,4株P(guān)EDV山西分離株屬于第一群,與2010年以后國(guó)內(nèi)流行毒株(除AH-M、SQ2014)的親緣關(guān)系較近,與2010年以前中國(guó)流行毒株、2個(gè)日本株、7個(gè)韓國(guó)株、2個(gè)疫苗株的親緣關(guān)系較遠(yuǎn)。研究結(jié)果提示山西省流行的PEDV發(fā)生較明顯的變異,需研發(fā)新的疫苗來(lái)控制PEDV的暴發(fā)。
