陸地棉光敏雄性不育基因ys-1的遺傳分析與初步定位

【摘要】:【目的】精細定位和克隆陸地棉光敏雄性不育基因!痉椒ā繌年懙孛拗040029的航天誘變后代中選育出新型光敏雄性不育系中9106,以中9106與11個陸地棉材料配制雜交組合,初步分析了中9106的雜種優(yōu)勢。以中9106為母本與陸地棉樂土603構(gòu)建遺傳分離群體F1、F2,分析不育性狀的遺傳特征。利用集團分離分析法篩選簡單序列重復(Simple sequence repeat,SSR),并在包含186個單株的中9106×樂土603 F2群體中用上述SSR初步定位不育基因!窘Y(jié)果】中9106為母本×陸地棉樂土603的F1單株育性均表現(xiàn)正常,而F2群體中的可育單株數(shù)∶不育單株數(shù)符合3∶1的分離比,推測中9106的不育性狀受1對隱性核基因控制。利用集團分離分析法從16 544對SSR中篩選到18對與該不育基因連鎖的標記。利用中9106×樂土603的F2群體和18對SSR標記,將不育基因定位于D12染色體,位于標記NAU3442與CGR6339之間,遺傳距離均為0.2c M,將該不育基因命名為ys-1!窘Y(jié)論】本研究有助于ys-1的精細定位及其克隆。
[Abstract]:[objective] to localize and clone the photoperiod sensitive male sterile gene of upland cotton. [methods] A new type of Guang Min male sterile line, 9106, was selected from 040029 space mutagenetic progenies of upland cotton, and the hybrid combinations were prepared between Zhong9106 and 11 upland cotton materials. The heterosis of Zhong 9106 was preliminarily analyzed. The genetic segregation populations F1 and F2 were constructed with Zhong 9106 as female parent and Letu 603 of upland cotton, and the genetic characteristics of sterile traits were analyzed. Simple sequence repeat (Simple sequence repeat,SSR was screened by group separation analysis), The sterile genes were initially located by SSR in 9106 脳 Letu 603 F2 population containing 186 plants. [results] the fertility of F1 plants with 9106 female parent 脳 upland cotton 603was normal. However, the number of fertile plants in F2 population: the number of sterile plants per plant was in accordance with the segregation ratio of 3:1, and it was inferred that 9106 of the sterile traits were controlled by a pair of recessive nuclear genes. 18 pairs of markers linked to the sterile gene were screened from 16544 pairs of SSR by group segregation analysis. Using 9106 脳 Letu 603F2 population and 18 pairs of SSR markers, the sterile gene was located on chromosome D12, between marker NAU3442 and CGR6339, and the genetic distance was 0.2cm. Naming the sterile gene ys-1. [conclusion] this study is helpful to the fine mapping and cloning of ys-1.
