RNAi沉默PTHrP基因的表達(dá)對SPC-A-1BM細(xì)胞侵襲力影響的研究

【摘要】:目的:探討甲狀旁腺激素相關(guān)蛋白(parathyroid hormone-related protein,PTHr P)基因的沉默對SPC-A-1BM細(xì)胞侵襲能力及其對細(xì)胞核因子κB受體活化因子配體(receptor activator of nuclear factor-κB ligand,RANKL)基因表達(dá)的影響。方法:本實(shí)驗(yàn)共設(shè)置5個(gè)組,其中1-3組為實(shí)驗(yàn)組(小si RNA干擾組,針對目的PTHr P基因設(shè)計(jì)3條不同序列的si RNA-PTHr P),4組:陰性對照組(非特異性陰性對照si RNA),5組:空白對照組(無si RNA干擾)。通過核酸轉(zhuǎn)染試劑脂質(zhì)體Lipofectamine TM2000(Lipo2000)介導(dǎo)小干擾si RNA轉(zhuǎn)染高轉(zhuǎn)移性人肺腺癌細(xì)胞株(SPC-A-1BM)細(xì)胞;Transwell細(xì)胞遷移實(shí)驗(yàn)用于檢測轉(zhuǎn)染后SPC-A-1BM細(xì)胞的侵襲能力;實(shí)時(shí)熒光定量聚合酶鏈反應(yīng)(Real-time quantitative polymerase chain reaction,Real-time PCR)用于檢測轉(zhuǎn)染后PTHr P基因的RNA表達(dá)水平;Western-Blot方法檢測轉(zhuǎn)染后PTHr P及RANKL的表達(dá)水平。結(jié)果:轉(zhuǎn)染12小時(shí)后進(jìn)行的Transwell試驗(yàn),顯示各組穿過Transwell人工膜并附著于上層小室底面的細(xì)胞,分別進(jìn)行細(xì)胞計(jì)數(shù),并進(jìn)行統(tǒng)計(jì)分析。結(jié)果顯示PTHr P特異性轉(zhuǎn)染組(1組、2組、3組)與對照組(4組、5組)相比,穿過人工膜的數(shù)量明顯減少,抑制率分別為63.8%、60.3%、66.0%,經(jīng)單因素方差分析,LSD法組間兩兩比較,結(jié)果顯示,實(shí)驗(yàn)組1、實(shí)驗(yàn)組2、實(shí)驗(yàn)組3均和對照組4組及5組的差異有統(tǒng)計(jì)學(xué)意義(P0.01),4組與5組間無明顯差異(P0.05);Real-time PCR實(shí)驗(yàn)顯示,實(shí)驗(yàn)組1組、2組及3組的PTHr P基因的表達(dá)顯著被抑制,抑制率分別為72%、75%、73%(P0.01),而對照組4組與5組之間對比,PTHr P基因的表達(dá)并沒有統(tǒng)計(jì)學(xué)差異;Western-Blot實(shí)驗(yàn)顯示,對于PTHr P蛋白,實(shí)驗(yàn)組1組、2組、3組的條帶灰度值之間無統(tǒng)計(jì)學(xué)差異(P0.05);4組、5組條帶之間無統(tǒng)計(jì)學(xué)差異(P0.05);無論1組、2組還是3組,它們的條帶灰度值與4組、5組對比,差異均具有統(tǒng)計(jì)學(xué)意義(P0.01)。而對于RANKL蛋白而言,同樣的,1組、2組、3組的條帶灰度值之間無統(tǒng)計(jì)學(xué)差異,(P0.05);4組、5組條帶之間無統(tǒng)計(jì)學(xué)差異,(P0.05);無論1組、2組還是3組,它們的條帶灰度值與4組、5組對比,差異均具有統(tǒng)計(jì)學(xué)意義(P0.01)。結(jié)論:利用RNAi技術(shù)轉(zhuǎn)染SPC-A-1BM細(xì)胞沉默PTHr P基因可明顯抑制腫瘤細(xì)胞的侵襲能力,并能減少RANKL蛋白的表達(dá);本實(shí)驗(yàn)為PTHr P介導(dǎo)的骨轉(zhuǎn)移瘤基因沉默療法提供了理論基礎(chǔ),肺癌骨轉(zhuǎn)移的治療提供了新的治療靶點(diǎn)。
