馬尾松PmCY P77A 1基因的克隆及表達分析

【摘要】:在進行馬尾松抗蟲材料轉(zhuǎn)錄組測序分析時發(fā)現(xiàn)一個編碼CYP77A1的cDNA,命名為PmCYP77A1。序列分析結(jié)果顯示,該基因全長1 620 bp,編碼539個氨基酸。氨基酸序列分析表明其具有CYP450家族基因的保守結(jié)構(gòu)域,但CYP77A1基因中的FXXGXXXCXG保守特征序列最后一個堿基被A堿基所代替。與其他植物核酸序列進行比對覆蓋率和同源性均較差,而氨基酸序列比對覆蓋率達到88%~96%,但同源性不高,基本都在50%~55%之間,PmCYP77A1與白云杉遺傳聚類最近,與其他被子植物遺傳聚類較遠。利用熒光定量PCR技術(shù)分析得知,PmCYP77A1在馬尾松的不同組織中均有表達,其中在嫩莖、雌雄花和嫩葉中的表達量較高,而在球果、成熟組織和根系中的表達量最低。在不同抗蟲材料日變化中PmCYP77A1表達量出現(xiàn)先升高后降低的表達趨勢,該基因在抗蟲材料中表達啟動早。研究結(jié)果將為進一步研究PmCYP77A1的生物學功能及在馬尾松抗蟲防御中的作用機理奠定基礎(chǔ)。

The results of sequence analysis showed that the full length of the gene was 1620 bp, encoding 539 amino acids.
