【摘要】:【目的】對(duì)中華按蚊(Anopheles sinensis)基因組中的乙酰膽堿酯酶(Ace)基因進(jìn)行鑒定和生物信息學(xué)分析!痉椒ā客ㄟ^(guò)同源性搜索得到1條乙酰膽堿酯酶(Ace)基因序列,命名為AsAce2,分析了該基因的基因結(jié)構(gòu)以及所編碼蛋白的理化性質(zhì)、亞細(xì)胞定位、二級(jí)結(jié)構(gòu)、三級(jí)結(jié)構(gòu)和系統(tǒng)發(fā)生關(guān)系!窘Y(jié)果】AsAce2基因全長(zhǎng)為4 008bp,編碼區(qū)序列長(zhǎng)度為1 941bp,共編碼647個(gè)氨基酸;AsAce2蛋白與其他蚊蟲(chóng)的Ace2蛋白具有較高的同源性,在系統(tǒng)發(fā)育關(guān)系上與岡比亞按蚊(Anopheles stephensi)的Ace2蛋白和不吉按蚊(Anopheles gambiae)的Ace2蛋白關(guān)系更近;AsAce2基因結(jié)構(gòu)中含有3個(gè)1位和2個(gè)2位內(nèi)含子;AsAce2蛋白分子式為C_(3222)H_(4925)N_(875)O_(947)S_(32),為親水性蛋白,蛋白定位在細(xì)胞周質(zhì),在13~32位氨基酸為跨膜區(qū),無(wú)疏水區(qū)和信號(hào)肽,二級(jí)結(jié)構(gòu)主要以不規(guī)則卷曲為主。【結(jié)論】豐富了相關(guān)基礎(chǔ)數(shù)據(jù),為進(jìn)一步研究該基因的功能奠定了基礎(chǔ)。
[Abstract]:[objective] to identify and bioinformatics analysis of acetylcholinesterase (Ace) gene in the genome of Anopheles sinensis (Anopheles sinensis). [methods] A sequence of acetylcholinesterase (Ace) gene named AsAce2, was obtained by homology search. The gene structure of the gene and the physical and chemical properties, subcellular localization, secondary structure, tertiary structure and phylogenetic relationship of the encoded protein were analyzed. [results] the full length of AsAce2 gene was 4 008bp, and the length of coding region was 1 941bp. the relationship between the gene structure and the physical and chemical properties, subcellular localization, secondary structure, tertiary structure and phylogeny of the gene was analyzed. A total of 647 amino acids were encoded. AsAce2 protein has high homology with Ace2 protein of other mosquitoes, and is more closely related to Ace2 protein of Anopheles gambiae (Anopheles stephensi) and Ace2 protein of Anopheles gambiae (Anopheles gambiae) in phylogenetic relationship. The structure of AsAce2 gene contains 3 1 loci and 2 2 introns. The molecular formula of AsAce2 protein is C3222 H4925 N875O947S32, which is hydrophilic protein.
