發(fā)菜RNA-seq轉(zhuǎn)錄組分析及耐旱相關(guān)基因篩選
[Abstract]:Nostoc is a kind of terrestrial nitrogen-fixing cyanobacteria, which has important ecological and resource value. However, the information of genome and transcript of lettuce is still lacking at present. In this study, the Illumina Hi Seq TM 2500 high throughput sequencing platform was used to analyze the transcripts of fully absorbed and dehydrated seaweed. A total of 30,013,560 clean reads,3 100 Unigene, sequences with an average length of 854 nt. were obtained by splicing and assembling. The Unigene sequence was compared with NR,NT,Swiss-Port,PFAM,GO,COG,KEGG (Evalue1.0E~ (- 5) database, and a total of 2874 Unigene were annotated. Through the classification of GO functions, 917 Unigene are mapped to different functional nodes of GO, and through the comparison of Unigene and COG database, the 514 Unigene with COG function can be divided into 20 categories. By KEGG pathway analysis, 1 220 coding genes were involved in 135 known metabolic pathways. It was suggested that some genes of metabolic pathways such as glutathione carotenoid starch sucrose and ABC transporter were up-regulated in dehydrated algae suggesting that these pathways are closely related to drought tolerance of lettuce. Therefore, these annotated information provide an important basis for the excavation of drought-tolerant genes in the future, and also provide an experimental basis for the further study of molecular biology of the lettuce.
