CRISPR-dCas9系統(tǒng)在基因表達調(diào)控中的最新研究進展

【摘要】:CRISPR-Cas9系統(tǒng)自問世以來一直被廣泛地應用于基因組編輯技術(shù)中,直到人們通過點突變的方式將Cas9的Ruv C和HNH剪切結(jié)構(gòu)域失活得到d Cas9后,這個系統(tǒng)又有了新的功能——調(diào)節(jié)基因的表達水平。研究者通過改變g RNA的結(jié)構(gòu),并與RNA結(jié)合蛋白配對,從而招募轉(zhuǎn)錄因子以調(diào)節(jié)基因的轉(zhuǎn)錄水平,達到影響基因表達的目的。本文主要討論了CRISPR-d Cas9系統(tǒng)的結(jié)構(gòu)特點、作用原理和目前對調(diào)節(jié)基因表達的最新研究進展,以期為此領(lǐng)域的研究者提供參考。
[Abstract]:Since its inception, CRISPR-Cas9 system has been widely used in genome editing technology, until people inactivated the Ruv C and HNH splicing domains of Cas9 by point mutation to obtain d Cas9. The system has a new function-regulating the level of gene expression. By altering the structure of g-RNA and pairing with RNA-binding proteins, the researchers recruited transcription factors to regulate the transcriptional level of the gene to influence gene expression. In this paper, we mainly discuss the structure and mechanism of CRISPR-d Cas9 system, as well as the latest research progress in regulating gene expression, so as to provide references for researchers in this field.
