人溶菌酶基因在乳酸克魯維酵母中的表達(dá)

【摘要】:人溶菌酶是一種天然廣譜抑菌物質(zhì),在食品和醫(yī)藥工業(yè)有潛在應(yīng)用前景。為獲得高活性的人溶菌酶制劑,采用乳酸克魯維酵母表達(dá)系統(tǒng),對經(jīng)密碼子優(yōu)化的人溶菌酶基因(h LYZ)進(jìn)行分泌表達(dá)。將人工合成h LYZ插入到乳酸克魯維酵母表達(dá)載體p KLAC1,構(gòu)建重組載體p KLAC1-h LYZ,并用電脈沖法將SacⅡ線性化的重組質(zhì)粒轉(zhuǎn)化到乳酸克魯維酵母GG799中。通過全細(xì)胞PCR鑒定,最后獲得了一株多拷貝整合的基因工程菌h LYZ1。工程菌可以分泌表達(dá)分子量約14 ku的目的蛋白質(zhì),與預(yù)期大小相符。搖瓶發(fā)酵培養(yǎng)128 h,酶活最高達(dá)到1430 U/mL。抗菌活性檢測結(jié)果顯示,重組人溶菌酶對溶壁微球菌、大腸桿菌和枯草芽孢桿菌有較好的溶菌活性。本研究成功地在乳酸克魯維酵母中表達(dá)了重組人溶菌酶,表達(dá)的蛋白具有較高的酶活性,試驗結(jié)果為利用乳酸克魯維酵母表達(dá)系統(tǒng)規(guī);a(chǎn)重組人溶菌酶奠了基礎(chǔ)。
