共培養(yǎng)沙門菌對(duì)糞腸球菌N41株生長(zhǎng)特性及毒力基因的影響

【摘要】:為探討共培養(yǎng)條件下沙門菌對(duì)糞腸球菌N41分離株生長(zhǎng)特性及其不同生長(zhǎng)時(shí)期26種毒力基因轉(zhuǎn)錄的影響,將N41菌株、沙門菌菌株單獨(dú)/混合培養(yǎng)后,分別進(jìn)行菌落計(jì)數(shù)并繪制生長(zhǎng)曲線,觀察其生長(zhǎng)狀況及影響,同時(shí)提取其對(duì)數(shù)中期、對(duì)數(shù)后期、穩(wěn)定前期和穩(wěn)定期的總RNA,SYBR GreenⅠ實(shí)時(shí)熒光定量PCR方法分析N41菌株26種毒力基因的轉(zhuǎn)錄情況。結(jié)果顯示,共培養(yǎng)后沙門菌和N41生長(zhǎng)均受到影響,其中,在N41菌體濃度降低約3.9倍,而沙門菌菌體濃度降低約110倍。在N41菌株26種毒力基因中,沙門菌促進(jìn)了ebpA、ebpC、rnjB、ace、ebpR、psr等12種毒力基因在4個(gè)不同生長(zhǎng)時(shí)期的轉(zhuǎn)錄,但同時(shí)也降低了毒力基因SlyA和sprE在4個(gè)生長(zhǎng)期中轉(zhuǎn)錄水平;另外,除了CylL-S、CylL-L、efaA和AS在4個(gè)生長(zhǎng)時(shí)期變化不明顯外,其余的大部分毒力基因都在對(duì)數(shù)后期轉(zhuǎn)錄水平有明顯上升,但在其他生長(zhǎng)時(shí)期則表現(xiàn)不同。這說明,兩菌共培養(yǎng)后,N41菌株明顯抑制了沙門菌的生長(zhǎng),同時(shí)沙門菌也整體促進(jìn)了N41菌株毒力基因的轉(zhuǎn)錄。本試驗(yàn)為研究細(xì)菌間相互作用機(jī)制以及腸球菌的致病機(jī)制奠定基礎(chǔ)。
