【摘要】：為研究三角帆蚌HcTyr基因與內(nèi)殼色性狀的相關(guān)性,本實(shí)驗(yàn)根據(jù)已分離的三角帆蚌HcTyr基因進(jìn)行引物設(shè)計(jì)和片段擴(kuò)增,并用144只三角帆蚌對(duì)其多態(tài)性進(jìn)行篩選和驗(yàn)證,卡方檢驗(yàn)分析了SNP位點(diǎn)和三角帆蚌內(nèi)殼色相關(guān)性,并對(duì)HcTyr基因進(jìn)行了圖譜定位。結(jié)果顯示,在HcTyr基因上篩選出8個(gè)SNP位點(diǎn),其中有7個(gè)SNP位點(diǎn)與三角帆蚌內(nèi)殼色L、a及d E呈顯著相關(guān)性,用這7個(gè)SNP位點(diǎn)做單倍型構(gòu)建和分析,發(fā)現(xiàn)Ⅱ、Ⅲ及Ⅳ這3種單倍型在白色群體中出現(xiàn)的頻率極顯著高于在紫色群體中出現(xiàn)的頻率,而Ⅴ和Ⅶ這2兩種單倍型在紫色群體中出現(xiàn)的頻率極顯著高于白色群體。進(jìn)一步在商業(yè)養(yǎng)殖群體中驗(yàn)證發(fā)現(xiàn),Ⅳ和Ⅶ這2種單倍型可分別作為白色和紫色群體的優(yōu)勢(shì)單倍型。研究表明,三角帆蚌HcTyr基因可作為內(nèi)殼色相關(guān)的候選基因,其部分SNP位點(diǎn)可用于三角帆蚌分子輔助育種。另外本實(shí)驗(yàn)還將HcTyr基因定位在三角帆蚌LG16連鎖群上,這為進(jìn)一步解析該基因調(diào)控珍珠顏色形成的分子機(jī)制奠定了基礎(chǔ)。
[Abstract]:In order to study the correlation between HcTyr gene and color traits of inner shell of Hyriopsis cumingii, primer design and fragment amplification were carried out according to the isolated HcTyr gene of Hyriopsis cumingii, and its polymorphism was screened and verified with 144 Hyriopsis cumingii. The correlation between SNP locus and inner shell color of Hyriopsis cumingii was analyzed by Chi-square test, and the mapping of HcTyr gene was carried out. The results showed that eight SNP loci were screened on the HcTyr gene, among which 7 SNP loci were significantly correlated with the inner shell color L, a and d E of Hyriopsis cumingii.
