三角帆蚌HcTyr基因內(nèi)殼色性狀相關(guān)SNP篩選及圖譜定位

【摘要】:為研究三角帆蚌HcTyr基因與內(nèi)殼色性狀的相關(guān)性,本實驗根據(jù)已分離的三角帆蚌HcTyr基因進行引物設(shè)計和片段擴增,并用144只三角帆蚌對其多態(tài)性進行篩選和驗證,卡方檢驗分析了SNP位點和三角帆蚌內(nèi)殼色相關(guān)性,并對HcTyr基因進行了圖譜定位。結(jié)果顯示,在HcTyr基因上篩選出8個SNP位點,其中有7個SNP位點與三角帆蚌內(nèi)殼色L、a及d E呈顯著相關(guān)性,用這7個SNP位點做單倍型構(gòu)建和分析,發(fā)現(xiàn)Ⅱ、Ⅲ及Ⅳ這3種單倍型在白色群體中出現(xiàn)的頻率極顯著高于在紫色群體中出現(xiàn)的頻率,而Ⅴ和Ⅶ這2兩種單倍型在紫色群體中出現(xiàn)的頻率極顯著高于白色群體。進一步在商業(yè)養(yǎng)殖群體中驗證發(fā)現(xiàn),Ⅳ和Ⅶ這2種單倍型可分別作為白色和紫色群體的優(yōu)勢單倍型。研究表明,三角帆蚌HcTyr基因可作為內(nèi)殼色相關(guān)的候選基因,其部分SNP位點可用于三角帆蚌分子輔助育種。另外本實驗還將HcTyr基因定位在三角帆蚌LG16連鎖群上,這為進一步解析該基因調(diào)控珍珠顏色形成的分子機制奠定了基礎(chǔ)。
[Abstract]:In order to study the correlation between HcTyr gene and color traits of inner shell of Hyriopsis cumingii, primer design and fragment amplification were carried out according to the isolated HcTyr gene of Hyriopsis cumingii, and its polymorphism was screened and verified with 144 Hyriopsis cumingii. The correlation between SNP locus and inner shell color of Hyriopsis cumingii was analyzed by Chi-square test, and the mapping of HcTyr gene was carried out.
