越橘類黃酮化合物轉(zhuǎn)運(yùn)相關(guān)基因MATE的克隆與表達(dá)分析

【摘要】:植物中多藥和毒性化合物外排(MATE)轉(zhuǎn)運(yùn)蛋白參與類黃酮在液泡中積累的過程。以高叢越橘品種"北衛(wèi)"為試驗(yàn)材料,應(yīng)用染色體步移和cDNA末端快速克隆技術(shù)克隆MATE轉(zhuǎn)運(yùn)蛋白基因的cDNA全長序列,命名為VcMATE6,序列提交到GenBank,登錄號為KF875437。序列分析表明VcMATE6的cDNA全長1 557 bp,編碼518個(gè)氨基酸,推測的蛋白相對分子質(zhì)量為56.39 ku,理論等電點(diǎn)為6.12。多重序列比對顯示VcMATE6與MATE型類黃酮轉(zhuǎn)運(yùn)蛋白有較高的同源性。聚類分析表明VcMATE6與MtMATE2、MlMTP77、AtTT12、VvAM1等一類植物典型的MATE型類黃酮轉(zhuǎn)運(yùn)蛋白聚為一類。VcMATE6具有2個(gè)MATE家族特有的Mat E結(jié)構(gòu)域,TMHMM在線軟件預(yù)測VcMATE6含有12個(gè)跨膜螺旋。實(shí)時(shí)熒光定量PCR分析表明,VcMATE6基因在越橘各器官和果實(shí)發(fā)育各階段均有表達(dá),但表達(dá)量存在差異,藍(lán)果種子中表達(dá)量最高,極顯著高于其他器官以及藍(lán)果的果皮和果肉組織,其次為根,果實(shí)發(fā)育過程中VcMATE6表達(dá)量隨果實(shí)成熟度表現(xiàn)出增加的趨勢。
[Abstract]:The efflux of (MATE) transporter from plant multidrug and toxic compounds is involved in the accumulation of flavonoids in vacuoles. The full-length cDNA sequence of MATE transporter gene was cloned by chromosomal walking and rapid cloning of cDNA terminal using "Beiwei", a variety of blueberry cultivars with high density, named VcMATE6, sequence and GenBank, accession number KF875437.. Sequence analysis showed that the cDNA length of VcMATE6 was 1 557 bp, encoding 518 amino acids, and the predicted molecular weight of the protein was 56. 39 ku, with a theoretical isoelectric point of 6. 12. Multiple sequence alignment showed high homology between VcMATE6 and MATE type flavonoid transporter. Cluster analysis showed that some typical MATE flavonoid transporters, such as VcMATE6 and MtMATE2,MlMTP77,AtTT12,VvAM1, were clustered into a class. VcMATE6 had two Mat E domains unique to the MATE family, and TMHMM online software predicted that VcMATE6 contained 12 transmembrane helices. Real-time fluorescence quantitative PCR analysis showed that VcMATE6 gene was expressed in all organs and fruit development stages of blueberry, but there were differences in the expression levels. The highest expression level of VcMATE6 gene was found in blueberry seeds, which was significantly higher than that in other organs and the peel and pulp tissues of blueberry. The second was root, and the expression of VcMATE6 increased with fruit maturity during fruit development.
