短時熱激后家蠶對核型多角體病毒的抗性及相關(guān)抗菌肽和熱激蛋白基因表達水平的變化

【摘要】:【目的】探討短時熱激對家蠶Bombyx mori抵抗核型多角體病毒能力的影響,為深入研究家蠶熱激響應(yīng)的抗病毒免疫機制提供參考!痉椒ā考倚Q5齡幼蟲42℃熱激15 min后,經(jīng)口喂食家蠶核型多角體病毒(Bombyx mori nucleopolyhedrovirus,Bm NPV)(5.4×106PIBs/頭)后正常飼養(yǎng),觀察熱激對家蠶感染病毒的影響;同時,利用實時熒光定量RT-PCR技術(shù)檢測熱激后7類抗菌肽主基因(Bmcec D,Bmmor,Bmglve2,Bmdefe B,Bmatta1,Bmenbo2和Bmlebo3)以及6種熱激蛋白基因(Bmhsp90,Bmhsp70,Bmhsp40,Bmhsp19.9,Bmhsp21.4和Bmhsp25.4)在家蠶5齡幼蟲中腸組織中的表達變化;利用MEGA 5.0軟件對家蠶所有熱激蛋白進行系統(tǒng)進化分析!窘Y(jié)果】與未進行熱激處理相比,短時42℃熱激能夠明顯提高感染Bm NPV的家蠶5齡幼蟲的存活率。實時熒光定量RT-PCR結(jié)果表明,與未熱激處理相比,熱激處理后感染Bm NPV的家蠶5齡幼蟲中腸組織中Bmglve2和Bmhsp25.4的表達量顯著上升。另外,系統(tǒng)進化分析顯示,小分子熱激蛋白Bm HSP25.4在進化上特殊,單獨與大分子熱激蛋白(Bm HSP90,Bm HSP70和Bm HSP40)聚為一類!窘Y(jié)論】短時熱激能夠提高家蠶對Bm NPV的抗性,熱激蛋白基因Bmhsp25.4和抗菌肽基因Bmglve2可能在家蠶熱激響應(yīng)的抗病毒免疫中發(fā)揮關(guān)聯(lián)功能。
