干旱脅迫棉花轉錄組DNA損傷修復相關基因的分析

【摘要】:【目的】研究干旱脅迫下,棉花DNA損傷修復基因的表達情況,從整體水平探討棉花DNA損傷修復相關基因表達與抗旱的相關性。【方法】用2.5%PEG6000處理棉花幼苗,利用RNA-Seq技術對干旱脅迫下棉花幼苗的轉錄組進行測序!窘Y果】從棉花干旱脅迫響應的轉錄組中,篩選獲得差異表達的DNA損傷修復相關基因共51個,其中差異表達的上調基因23個,差異表達的下調基因28個,干旱脅迫能夠影響棉花DNA損傷修復相關基因的表達。選取4個差異基因進行生物信息學分析及qRT-PCR驗證,HMGB1、rec A1、UDGs和GMP synthase基因的表達變幅有一定的差異,但基因的表達趨勢一致,棉花轉錄組測序結果通過qRTPCR驗證是可靠的!窘Y論】DNA損傷修復相關基因可能與干旱脅迫有一定的相關性,干旱脅迫能夠影響棉花DNA損傷修復相關基因的表達。
[Abstract]:[objective] to study the expression of DNA damage repair genes in cotton under drought stress, and to explore the correlation between the expression of DNA damage repair genes and drought resistance in cotton seedlings. [methods] Cotton seedlings were treated with 2.5%PEG6000. The transcriptome of cotton seedlings under drought stress was sequenced by RNA-Seq technique. [results] 51 differentially expressed genes related to DNA damage repair were screened from the transcriptional groups of cotton drought stress response. Among them, 23 genes were up-regulated and 28 genes were down-regulated. Drought stress could affect the expression of genes related to DNA damage and repair in cotton. Four differentially expressed genes were selected for bioinformatics analysis and qRT-PCR verification. There were some differences in the expression of HMGB1,rec A1UDGs and GMP synthase genes, but the trend of gene expression was the same. [conclusion] DNA damage repair related genes may be related to drought stress, drought stress can affect the expression of cotton DNA damage repair related genes.
