葡萄VvCOL5基因克隆及在胚珠發(fā)育過(guò)程中的表達(dá)分析

【摘要】:以葡萄無(wú)核品種無(wú)核白和有核品種黑比諾為材料,克隆葡萄CONSTANS基因家族的1個(gè)成員并將其命名為VvCOL5基因,分析基因結(jié)構(gòu),進(jìn)行染色體定位和系統(tǒng)發(fā)育樹(shù)的構(gòu)建,檢測(cè)VvCOL5基因在無(wú)核和有核葡萄胚珠(幼種子)發(fā)育過(guò)程的表達(dá)變化。結(jié)果表明:VvCOL5基因cDNA長(zhǎng)度為1 374bp,包含有2個(gè)外顯子和1個(gè)內(nèi)含子結(jié)構(gòu),并含有2個(gè)B-box結(jié)構(gòu)域和1個(gè)CCT結(jié)構(gòu)域,屬于CO家族的第一類,在GenBank登錄號(hào)為KX077245。在受精后胚珠(幼種子)發(fā)育過(guò)程中,VvCOL5基因在無(wú)核白與黑比諾中呈現(xiàn)出了相反的表達(dá)模式。其無(wú)核白受精后10~30d其表達(dá)緩慢上調(diào),30~35d維持在較高水平,35~40d緩慢下降,后在40~45d又快速上調(diào)到較高水平;而在黑比諾中10~20d下降,在20~35d之間相對(duì)平穩(wěn),35~40d緩慢上升,最后40~45d時(shí)又下降到較低水平。無(wú)核白中VvCOL5基因在各時(shí)期的表達(dá)量均高于黑比諾,可見(jiàn)VvCOL5基因在無(wú)核和有核品種中表達(dá)差異較大,其與葡萄種子發(fā)育有一定關(guān)系。
[Abstract]:A member of grape CONSTANS gene family was cloned from seedless grape varieties denuclearized and Pinot Noir, named VvCOL5 gene, gene structure was analyzed, chromosome mapping and phylogenetic tree construction were carried out. To detect the expression of VvCOL5 gene during the development of seedless and nucleated grape ovule (young seeds). The results show that the length of cDNA of VvCOL5 gene is 1 374 BP, including 2 exons and 1 intron structure, 2 B-box domains and 1 CCT domain, which belong to the first class of CO family, and the GenBank accession number is KX077245.. During the development of ovules (young seeds) after fertilization, the VvCOL5 gene expressed in denuclearized and Pinot Noir. The expression increased slowly at 10 ~ 30 days after fertilization, maintained at a higher level at 30 ~ 35 days, decreased slowly at 35 ~ 40 days, and then increased to a higher level at 40 ~ 45 days after fertilization. In Pinot Noir, it decreased at 10 ~ 20 days, remained stable at 20 ~ 35 days, increased slowly at 35 ~ 40 days, and then decreased to a lower level at 40 ~ 45 days. The expression of VvCOL5 gene in denuclearized white was higher than that in Pinot noir at every stage. It was found that the expression of VvCOL5 gene in seedless and nucleated varieties was different and had a certain relationship with grape seed development.
