馬藺DFR基因的克隆及生物信息學(xué)特征分析

【摘要】:以馬藺花瓣為試材,通過轉(zhuǎn)錄組測序,應(yīng)用PCR技術(shù)克隆馬藺花青素生物合成關(guān)鍵酶二羥黃酮醇-4-還原酶(dihydvroflavonol-4-reductase,DFR)基因并獲得其生物信息學(xué)特征。結(jié)果表明:馬藺DFR基因全長1 427bp,共編碼357個氨基酸(該基因命名為IlDFR,GenBank登錄號為KY907171)。該基因在氨基酸水平上與多種植物的DFR具有較高的同源性,在系統(tǒng)進(jìn)化上與同屬的荷蘭鳶尾(Iris hollandica)的DFR親緣關(guān)系較近;預(yù)測其蛋白質(zhì)相對分子質(zhì)量為39.99kDa,等電點為5.89,主要由α-螺旋和β-折疊構(gòu)成,定位于細(xì)胞質(zhì),屬于酸性親水不含信號肽的不穩(wěn)定類蛋白質(zhì)。馬藺IlDFR有一段氨基酸序列‘VTGASGYVGSWLVMKLLRDGY’與大部分物種的DFR特有相對保守的NADPH結(jié)合域非常相似,只有4個氨基酸殘基有所差異。
[Abstract]:The key enzyme of Iris anthocyanin biosynthesis, dihydroflavone alcohol 4-reductase (dihydvroflavonol-4-reductase,DFR), was cloned by PCR and its bioinformatics characteristics were obtained. The results showed that the total length of DFR gene was 1 427 BP, encoding 357 amino acids (IlDFR,GenBank accession number was KY907171). This gene has high homology with DFR of many plants at amino acid level, and is closely related to DFR of (Iris hollandica) of the same genus in phylogeny. The predicted molecular weight of the protein is 39.99 kDa and the isoelectric point is 5.89. The protein is mainly composed of 偽 -helix and 尾 -fold, located in the cytoplasm, and belongs to the unstable protein with acidic hydrophilic and no signal peptide. IlDFR has a segment of amino acid sequence 'VTGASGYVGSWLVMKLLRDGY', which is similar to the relatively conserved NADPH binding domain specific to DFR in most species, with only 4 amino acid residues being different.
